益气固本胶囊靶向调控miRNA-34a调节Treg/Th17免疫失衡治疗儿童哮喘的机制研究

免疫失衡是哮喘重要的发病机制之一，与气道炎症和气道重塑等病理变化密切相关。最新研究表明miRNA-34a靶向调控Foxp3、Notch1表达参与调节哮喘Treg/Th17免疫失衡。前期研究表明益气固本胶囊可有效减少哮喘病反复发作，具有调节Th1/Th2免疫失衡及增加Treg细胞数量的作用，故提出益气固本胶囊通过靶向调控miRNA-34a调节Treg/Th17免疫失衡治疗儿童哮喘的假说。本项目运用流式细胞术、RT-PCR、ELISA、Western-Blot等方法，分别研究益气固本胶囊调控miRNA-34a介导的Foxp3表达，以及miRNA-34a激活的Notch信号通路，调节Treg/Th17免疫失衡减轻气道炎症及抑制气道重塑的作用机制，阐释基于中医“治未病”理论，以及祛除“夙根伏痰”治法的益气固本胶囊治疗儿童哮喘的生物学机制，为传承国医大师王烈教授学术经验并在临床推广应用提供实验依据。

Immune imbalance is one of the important pathogenesis of asthma, which is closely related to pathologic changes such as airway inflammation and airway remodeling. Recent studies have shown that miRNA-34a targeted regulation of Foxp3 and Notch1 expression is involved in regulating the immune imbalance of Treg/Th17 in asthma. Previous studies have shown that Yiqi-Guben Capsule can effectively reduce recurrent asthma attacks, regulate Th1/Th2 immune imbalance and increase the number of Treg cells. Accordingly, this program proposes that Yiqi-Guben Capsule could treat childhood asthma by regulating Treg/Th17 immune imbalance through targeted regulation of miRNA-34a. It examines the regulation of Yiqi-Guben Capsule on miRNA-34a-mediated Foxp3 expression and the regulation of Treg/Th17 immune imbalance to reduce airway inflammation; besides, it studies the regulation of miRNA-34a-activated Notch signaling pathway and the regulation of Treg/Th17 immune imbalance to inhibit airway remodeling. In this program, a combination of flow cytometry, ELISA, RT-PCR, Western-Blot, and other research techniques will be applied comprehensively to investigate the biological mechanism of Yiqi-Guben Capsule based on the theory of “cure no disease” in TCM, and eliminating “long-cherished root and phlegm” in treating asthma in children， providing experimental basis for inheriting the academic experience and clinical application of professor Wang Lie, a master of Chinese medicine.
