非洲猪瘟是由非洲猪瘟病毒(ASFV)引起的对全球养猪业危害极大的传染病。ASFV囊膜蛋白CD2v既是重要的保护性抗原又是毒力相关蛋白,其红细胞吸附功能有助于病毒在机体中广泛扩散,是影响病毒毒力的一个重要因素,然而目前CD2v蛋白红细胞吸附功能相关氨基酸位点仍不明确。本项目拟利用慢病毒系统构建稳定表达CD2v的细胞系,建立无病毒感染CD2v红细胞吸附实验平台,用于验证CD2v红细胞吸附功能。将稳定表达CD2v的鼠源细胞系免疫BALB/c小鼠制备单克隆抗体,从中筛选具有抑制红细胞吸附功能的单克隆抗体,通过噬菌体展示技术鉴定其结合的抗原表位。利用定点突变的方法验证相关氨基酸对CD2v红细胞吸附功能的影响,最终明确CD2v参与红细胞吸附的关键氨基酸位点。该研究有助于我们更加深入了解CD2v的生物学功能,为进一步研制ASFV疫苗及抗病毒药物提供更充实的理论指导,具有重要理论意义和潜在商业价值。
英文摘要
African swine fever is an important infectious disease caused by african swine fever virus (ASFV),which is extremely harmful to the global swine industry. Studies show that the envelope protein CD2v of ASFV plays an important role in virus transmission, immune protection and virulence enhancement. The hemadsorption(HAD) function of CD2v has a very important relationship with the virus spread in the body, and is an important factor affecting virulence,but its key functional sites are still unknown. In this project, a stable cell line expressing CD2v will be constructed by lentivirus system to verify the HAD function of CD2v. A stable mouse cell line expressing CD2v will be used to immunize mice to produce CD2v monoclonal antibody, and the antibodies with the ability of hemadsorption inhibition (HAI) will be screened by HAD test. Phage display technique will be used to identify the HAD related epitopes of CD2v. By site-directed mutation of CD2v, the key sites in CD2v that play the role of HAD will be identified. This result will help us understand the biological function of CD2v in depth and provide more substantial theoretical guidance for the further development of ASFV vaccines and antiviral drugs. It has important theoretical significance and potential commercial value.
