我们已发表的研究发现AD模型小鼠脑内异常高表达的RIN3蛋白可增加毒性C99的产生及tau磷酸化(pTau),与AD的发生发展密切相关。多项研究揭示RIN3上游非编码区rs10498633为AD风险位点,我们也发现该位点致AD风险升高,近期我们发现rs10498633G>T显著增强RIN3表达,但机制不清。生物信息学预测发现rs10498633所在序列可能为RIN3远端增强子,远端增强子主要通过结合转录因子(TF)调控启动子活性影响转录,分析rs10498633的TF结合情况显示,多种TF在rs10498633T和G间具有不同的结合倾向性。因此,我们推测此变异通过影响TF与增强子的结合,上调RIN3启动子的转录活性引起RIN3表达升高,进一步导致C99和pTau的增多而增加AD风险。本项目我们将通过体内外实验验证我们的假说,探讨rs10498633在AD发生、发展中的作用及可能机制。
英文摘要
Our published research found the abnormally upregulated RIN3 in the brain of AD mice could increase the production of toxic C99 and tau phosphorylation, which are closely related to the occurrence and development of AD. A number of studies revealed that rs10498633, located in the upstream non-coding region of RIN3, is an AD risk locus. We also found that this site increases the risk of AD, besides, rs10498633 G>T significantly enhances the expression of RIN3, but the mechanism is still unclear. Through bioinformatic analysis, we found that rs10498633 is located in a highly probable distal enhancer region of RIN3. The distal enhancer mainly affects transcription by binding transcription factor (TF) to regulate promoter activity. Analysis of rs10498633 TF binding sites showed that a variety of TFs have different binding propensities between rs10498633 T and G. Therefore, we speculate that rs10498633 affects the binding of TF and enhancer and upregulates the transcriptional activity of the RIN3 promoter to increase the expression of RIN3, which further leads to an increase in Aβ and phosphorylated tau, increasing the risk of AD. In this project, we will verify the hypothesis through in vivo and in vitro experiments, and explore the role and possible mechanism of rs10498633 in the occurrence and development of AD.
阿尔茨海默病(AD)是发病率最高的神经退行性疾病,主要病理表现为Aβ过度沉积和tau过度磷酸化(pTau),明确Aβ、pTau异常增多机制及潜在防治靶点是AD研究领域的热点和难点。我们前期发现AD小鼠脑内异常高表达的RIN3既可导致Aβ前体增多又可导致pTau增多,从一崭新的角度探讨了Aβ、pTau异常增多的机制。因此,探索RIN3异常上调的机制对于明确AD的发病机制及寻找防治靶点具有重要意义。. 研究提示RIN3基因自身的异常可导致RIN3表达水平异常上调。多个研究报道RIN3上游非编码区rs10498633 G>T为AD发生的高危因素,我们的研究也证明rs10498633 G>T与AD发生有关,并进一步通过eQTL分析发现rs10498633 G>T的改变可增强RIN3表达。明确rs10498633如何增加RIN3表达水平将帮助了解AD中Aβ、pTau过度沉积的原因。. 生信分析提示rs10498633可能位于RIN3增强子区,通过影响转录因子与增强子的结合,上调RIN3启动子转录活性增强RIN3表达。我们进一步通过实验鉴定出RIN3核心启动子区,证明了rs10498633所在区段为RIN3增强子区,并进一步通过实验证实rs10498633 G>T的改变可进一步增强RIN3的表达。. 进一步地,我们通过生信分析、luciferase、过表达敲减实验等筛选出可能介导rs10498633对RIN3转录调控的转录因子,并进一步在转基因小鼠模型中探讨了RIN3表达上调后的小鼠行为学和生物学改变。该项目进展顺利,已完成主要的项目计划研究内容,探讨了rs10498633在AD发生发展中的作用及可能机制。同时,在该项目的资助下,完成了帕金森病中致病基因CD200R1及致病microRNA hsa-miR-4639-5p的转录调控研究,发表SCI 3篇,其中JCR 1区、影响因子8分以上 2篇。
