湘西苗医刘氏小儿推拿流派“推五经”调节哮喘患儿免疫平衡的宿主-菌群共代谢机制

既有研究表明，推拿能调节哮喘缓解期患儿的免疫平衡，也能有效改善脾虚哮喘大鼠的肠道菌群紊乱及炎症状态。宿主-菌群共代谢产物是肠道菌群发挥生物学作用的重要组分，而色氨酸代谢-AhR通路是肠道菌群调控哮喘免疫的中心环节。本课题旨在从推拿干预“肠道菌群-共代谢产物-免疫状态”的角度，探讨湘西苗医刘氏小儿推拿流派“推五经”调节缓解期哮喘患儿免疫平衡的宿主-菌群共代谢机制。课题将采用16S rRNA高通量测序、GC-MS定量代谢组学、流式细胞术、Western Blot等技术，首先基于临床研究推拿对哮喘缓解期患儿肠道菌群多样性、细菌功能特征、宿主-菌群共代谢产物的影响，及与Th细胞亚群分化、特征性细胞因子、临床疗效等表型的相关性；再应用粪菌移植技术，模拟推拿对哮喘小鼠的影响，基于色氨酸代谢-AhR通路，进一步研究宿主-菌群共代谢机制参与推拿调节哮喘免疫平衡的因果证据，为推拿防治儿童哮喘提供科学依据。

The purpose of this study is to explore the host flora co metabolism mechanism of the Liu Family Five-Meridian Management with Miao Medicine Tuina in the West of Hunan Province in terms of Tuina intervention on "intestinal flora co metabolites immune state". In this study, 16S rRNA high-throughput sequencing, GC-MS quantitative metabolomics, flow cytometry, Western blot and other technologies will be used.
