人参具有“大补元气”的功效,但是其功效是如何发挥作用目前仍然没有得到系统阐述,因此,“大补元气”传统功效的科学诠释是当前人参现代研究中不可回避也是必须回答的首要问题。本课题基于“气血学说”,以人参对红细胞形态和功能的影响为切入点,建立体外红细胞氧化应激模型和体内以“劳则气耗”中医理论指导下的拟临床“气脱”“气虚”模型,针对不同人参提取物开展筛选和验证,在筛选出人参活性成分的基础上,开展人参对红细胞膜Band-3蛋白的调控作用研究,并确定其作用靶点,最终从人参活性成分结合Band-3蛋白胞质结构域调控蛋白复合物装配调节红细胞形态和功能的角度系统阐述人参发挥“大补元气”传统功效的物质基础和生物学机制,课题的开展为以人参为原料的新药开发、临床用药、适宜人群和大健康产品开发提供重要的理论依据和数据支撑。

Ginseng has the effect of invigorating primordial qi potently, but how to play its role has not been systematically revealed yet. Therefore, the scientific interpretation of the traditional effect of invigorating primordial qi potently is the most important issue that cannot be avoided and must be clarified in the current modern research of ginseng. Based on the theory of qi and blood and the effect of ginseng on the morphology and function of red blood cells, the erythrocyte oxidative stress model in vitro and the pseudo-clinical model of depletion of qi and qi deficiency under the guidance of the theory of overexertion renders qi consumed in vivo were established. Different ginseng extracts were screened and verified. On the basis of screening out the active components of ginseng, the regulatory effect of ginseng on erythrocyte membrane Band-3 protein was investigated. Besides, its action target was determined.Finally, from the point of view of the combination of ginseng active components and Band-3 protein cytoplasmic domain to regulate the assembly of protein complex and regulate the morphology and function of red blood cells, this study systematically reveals the material basis and biological mechanism of ginseng to the traditional effect of invigorating primordial qi potently. The development of this project provides important theoretical basis and data support for the development of new drugs. Moreover, it offers clinical medication, suitable population and large health products using ginseng as raw materials.
