很多OT百合品种有着浓郁的花香,释放出特征花香成分1,8-桉树脑等单萜挥发物。前期研究发现它们的野生百合亲本岷江百合、淡黄花百合和通江百合中释放出高含量的1,8-桉树脑及伴随单萜挥发物,表现出“桉树脑盒”挥发物释放特点,但其生物合成关键酶基因CNS的功能尚不清楚。我们从岷江百合转录组文库中筛选出了4个候选LrCNS基因,在此基础上,本项目将克隆LrCNS基因全长,通过生物信息学分析、基因表达模式分析、体外酶促反应和亚细胞定位进行目标基因筛选和功能分析;然后在淡黄花百合和通江百合中进行CNS基因同源克隆和功能验证。在释放“桉树脑盒”挥发物的百合品种中克隆CNS同源基因并分析其亲缘关系;在不释放“桉树脑盒”挥发物的东方百合品种中瞬时表达LrCNS基因后检测花香产物,研究百合品种中“桉树脑盒”挥发物形成的分子机制。本研究对我国野生百合的优异性状进行挖掘,为百合花香育种提供有力指导和参考。
英文摘要
Most Oriental × Trumpet (OT) lily cultivars have fragrant flowers and release 1,8-cineole and other monoterpenoids as characteristic components. Our previous studies have found that high levels of 1,8-cineole and several other monoterpenoid were released simultaneous from the flowers of Lilium regale, L. sulphureum and L. sargentiae, which are wild ancestors of the OT hybrids. This phenomenon is called as ‘cineole cassette’. The functional characterization 1,8-cineole synthase gene (CNS), which are responsible for the biosynthesis of ‘cineole cassette’ volatiles, is still unclear in Lilium and other monocot plants. Based on transcriptome data we have screened four candidate LrCNS genes from L. regale. In this study, after cloning the full-length of LrCNS, we will carry out bioinformatic analysis, spatial and temporal expression pattern analysis, in vitro enzymatic activity test and subcellular localization to identify the specific gene and its function. Then homologous CNS genes will be cloned from L. sulphureum and L. sargentiae and their functions will be characterized. The CNS genes will be cloned from lily cultivars that emitted ‘cineole cassette’ monoterpenoids then their genetic relationship will be analyze with wild lilies. After transient expression of LrCNS in Oriental lily cultivar which do not release ‘cineole cassette’ monoterpenoids, the floral volatiles will be detected and the molecular mechanism of ‘cineole cassette’ volatiles biosynthesis in lily cultivars will be discussed. This study focuses on the mechanism research of excellent characteristics from wild Lilium species, and will provide guidance and reference in the floral scent research of Lilium plants.
