云南省是满天星的主产区,但由于新品种培育滞后于生产,且不能满足市场需求,目前主要栽培国外的商业品种,这严重制约了我国满天星产业的发展。因此,培育满足市场需求的满天星新品种具有重要意义。然而,传统杂交育种方法受到减数分裂重组抑制基因的限制,杂交后代遗传重组率低,极大的制约了植物育种的效率和品质。满天星重组抑制基因及其调控重组的分子机理研究,可为通过提高减数分裂重组率对满天星进行创新育种奠定基础。为此,本研究拟对满天星基因组进行测序,鉴定和克隆满天星减数分裂重组抑制基因,利用TILLING技术筛选满天星重组抑制基因突变体,通过细胞学方法研究突变体减数分裂过程中染色体的行为特征,探索重组抑制基因调控满天星减数分裂重组的分子机理,为充分利用满天星基因潜力培育新品种提供新途径和思路。
英文摘要
Yunnan Province is the main producing area of Gypsophila paniculata in China, but the cultivation of new varieties lags behind production and couldn’t meet the market demand, the main cultivated G. paniculate was the foreign commercial varieties, which seriously restricts the development of domestic flower industry in G. paniculata. Therefore, it is of great significance to cultivate new varieties of G. paniculata which meet the market demand. However, traditional hybrid breeding is limited by meiotic recombination suppressors, resulting in low recombination rate of hybrid progeny, which greatly restricts the efficiency and quality of plant breeding. Study the meiotic recombination suppressors and its molecular regulation mechanism could lay the foundation for the innovative breeding of G. paniculata by increasing the meiotic recombination rate. To this end, this study intends to do the genome sequence of G. paniculata, identify and clone the meiotic recombination suppressors. Then, using the TILLING method to isolate the anti-CO mutants, and observe the chromosome behavioral characteristics of the mutants during meiosis through cytology approach, with the aim to explore the molecular regulation mechanism of recombinant suppressors during meiosis in G. paniculata. This study will provide new approaches and ideas for the effective use of gene potential to breed new varieties in G. paniculate.
