大豆是我国重要的粮食和油料作物。磷有效性低是酸性土壤上限制大豆生长和产量的重要因素之一。低磷胁迫不仅影响根瘤的生长,而且抑制根瘤生物固氮功能的发挥。近年来的研究表明,根瘤自身也进化出了一些低磷胁迫适应性机制,但是,关于其中的分子机理的研究较少。本项目在前期克隆了一个主要在根瘤中受低磷胁迫上调表达的MADS家族转录因子,GmAGL82,亚细胞定位分析表明GmAGL82定位于细胞核。本项目拟通过构建超量或干涉表达GmAGL82的大豆整株转化材料,研究GmAGL82调控大豆根瘤适应低磷胁迫的生理机制,并进一步结合转录组测序以及酵母单杂交分析,明确GmAGL82的分子调控途径,以期为酸性土壤上以磷促氮的大豆高效栽培措施提供理论基础。
英文摘要
Soybean is an important grain and oil crop in China. Low phosphorus availability is one of the most important factors limiting soybean growth and yield in acid soil. Phosphorus deficiency not only inhibits nodule growth, but also affects the function of symbiotic nitrogen fixation in soybean nodules. In recent years, studies have shown that nodules have evolved a series of adaptive mechanisms in response to phosphorus deficiency, but the molecular mechanism of which has not been well studied. Before this study, we cloned a MADS family transcription factor, GmAGL82, which was mainly up-regulated by phosphorus deficiency in nodules, and subcellular localization analysis showed that GmAGL82 was localized in the nucleus. The whole plant transformation materials with over- or interference expression of GmAGL82 can be constructed in the following study to analysis the physiological mechanism of GmAGL82 regulating soybean nodules to adapt to phosphorus deficiency. Furthermore, combining with transcriptome sequencing and yeast one-hybrid analysis, the molecular regulation pathway of GmAGL82 in nodules adaption to phosphorus deficiency can be elucidated. Our study will provide theoretical basis for high-efficiency cultivation measures of soybean on acid soil with phosphorus and nitrogen.
