中药药性理论是中医药学理论的重要组成部分,如何辨识、确认动物药咸寒药性的表达,是中药药性理论研究的难点之一。基于前期研究基础,提出科学假说:MAPK/JNK和PI3K/AKT交互串话相关蛋白组合变化是水蛭-地龙方“咸寒”药性表达在治疗缺血性脑卒中的分子生物学基础之一。以咸寒药性理论为指导,咸寒药性代表方(水蛭-地龙方)为探针,在初步明确水蛭-地龙方药效与MAPK/JNK和PI3K/AKT交互串话网络相关的基础上,应用数学建模方法,进一步确定中药组方整体药性特征和水蛭-地龙方与相关蛋白的紧密度,并通过非咸寒中药组方体内外反证,综合阐释MAPK/JNK和PI3K/AKT交互串话网络中相关蛋白组合与咸寒药性之间的关系,从而明确动物药咸寒药性表达,并建立“药效机理初探——数学模型聚焦——同效它性反证”的中药药性表达研究模式,为中药药性理论研究提供新思路、新策略。
英文摘要
The property theory of traditional Chinese medicine is an important part of the theory of traditional Chinese medicine. And how to identify and confirm the expression of saline-cold properties in animal medicine is one of the difficulties in the theoretical study of traditional Chinese medicine. Based on the previous research foundation, the scientific hypothesis that the related proteins combination changes of the crosstalk between MAPK/JNK and PI3K/AKT is one of the molecular biological basis for analyzing the expression of ‘saline-cold’ properties of Shuizhi-Dilong recipe in ischemic cerebral stroke treatment is proposed. Guided by the saline-cold traditional Chinese medicinal property theory, the representative salt-cold medicine (Shuizhi-Dilong recipe) was used as a probe to clear the efficiency and related proteins in the crosstalk network between MAPK/JNK and PI3K/AKT. Then the mathematical modeling method was used to further determine traditional Chinese medicinal properties of the prescription and its compactness with related proteins. Finally, it was proved by other prescriptions experiments in vivo and vitro to explain the relationship between proteins combination in the crosstalk network MAPK/JNK, PI3K/AKT and the saline-cold properties comprehensively, so as to clarify the expression of saline-cold properties in animal medicine, and establish the expression pattern of the property of traditional Chinese medicine that ‘explore the mechanism of drug efficacy primarily-mathematic model to focus-making counterevidence with the same effect and different property prescription’,which provides new ideas and strategies for the property research of traditional Chinese medicine.
