沙棘是青海省重要的生态经济林浆果植物资源,具有很高的开发利用价值。沙棘含有大量的维生素、黄酮类、生物碱类等营养和活性物质。其中,沙棘维生素p(Vp)是以生物类黄酮为主成分的有效物质群,具有增强毛细血管壁韧性、保护血管、降血脂等功效。深入研究沙棘化学成分及药理作用,对深度开发利用沙棘资源,发展地方经济具有很大的促进作用,符合青海省“生态立省”的发展战略。本项目前期研究了沙棘Vp提取物的制备,并进行了体外血管环实验、高血脂大鼠的血管内皮功能保护作用研究,明确了沙棘Vp血管保护的功能。在此基础上,本项目将深入开展沙棘Vp的化学成分分离、制备,通过沙棘Vp及单体成分对内皮细胞相关基因及蛋白表达的影响,筛选出沙棘Vp有效成分群,利用血管损伤大鼠模型,从分子及蛋白水平,从EMPs与eNOS/NO信号通路的关系探讨其作用靶点及作用机制。
英文摘要
As an important economic forest berry of Qinghai, Sea Buckthorn (Hippophae rhamnoides L.) has a big value to exploit. According to the recently research, Sea buckthorn not only contains abundant vitamins, fatty acids and traces elements, but also contains flavonoid and phenolic compounds. Sea buckthorn Vp were compound groups mainly made up by bioactive flavonoids, such as catechin, isorhamnetin,.quercetin, proanthocyanidins, and other flavonoids and phenolic compounds. Sea buckthorn Vp have antioxidant, anti-fatigue, antitumor, anti-aging, immune regulation, anti-diabetes, and hypolipidemic effect, and it can prevent cardiovascular disease. It will promote the local economy if study chemical composition and pharmacological activities of sea buckthorn in depth.. At the earlier stage of this subject, sea buckthorn Vp preparation and the activities on cardiovascular has been researched. This subject will study the chemical compounds of Vp, and obtain compounds with high purity, identify the structure, then test the effects of crude sample and monomer on HUVECs. Test and verify the protect function on HUVECs through mRNA and protein expressions of related genes. select the effective compounds group, than experimentation on animals. Based on MPs and eNOS/NO signal pathway to study protection against vascular endothelial function of Sea Buckthorn Vp, though provide a basis to further researches and exploit of Sea Buckthorn.
