英文摘要
Based on the literature and our preliminary data, we hypothesize that in the injured distal lung tissue, the alveolar epithelial type II progenitor cells (ATIICs) express a disease-specific miR-145-miR-371b-5p+exosome-miRNA (Exo-miR) fingerprint to promote the ATIIC-specific differentiation for the tissue regeneration. In this proposed project, we plan to transplant the genetically modified Lamp2b-GFP+ATIICs that can efficiently express and load the Lamp2b-GFP fusion peptide to their derived exosomes. Thus, we will be able to trace the Lamp2b-GFP+miR-145—miR-371b-5p+exosome-mediated intercellular communication in the injured microenvironment in vivo to explore the mechanism underlying the disease-specific Exo-miR mediated tissue regeneration. In addition, this is the first project to generate the ATIIC-specific Lamp2b-GFP+miR-145—miR-371b-5p+exosomes to explore their therapeutic potential to induce the endogenous ATIIC-specific differentiation for tissue regeneration, which we believe will open a novel avenue for the development of precision targeted therapies for severe lung injury.
肺远端组织损伤或疾病如肺纤维化以及特发性肺间质纤维化往往导致肺泡上皮II型干/祖细胞 (Alveolar epithelial type II stem/progenitor cell, ATIIC)的功能丧失,继而出现进行性、不可逆的肺泡组织结构破坏,严重者会导致肺部肿瘤的发生。在本项目的大力支持下,我们成功构建了可分泌Lamp2b-GFP+外泌体的hiPSC-ATIIC,并分析疾病特异性miR-145-miR-371b-5p+Exo-miR 调节ATIIC 定向分化、增殖潜能。另外,我们还明确了ATIIC来源的外泌体miRNA信号在控制ATIIC特异性增殖或分化的分子机制。在另外一项扩展性研究中,我们发现去泛素化酶DUB3可通过MAGEA3-P53信号通路驱动肝癌的进展,提示抑制DUB3的去泛素化酶活性可能成为一种潜在治疗HCC的策略和方案。本项目的深入实施可能为包括严重肺损伤和肝癌肿瘤患者的治疗提供全新的方案和策略。
