Our study found that HOXB9 plays an important role in the invasion and metastasis of hepatocellular carcinoma (HCC), but the regulatory mechanism of HOXB9 has not been fully elucidated. Previous studies have found that eEF1A1 plays an important role in the progression of malignant tumors (Cancer research,2016). In our previous study, some phenomenon were found: eEF1A1 was overexpressed in HCC tissues and positively correlated with HOXB9 expression; Knockdown of eEF1A1 expression can down-regulate HOXB9 expression and decrease the invasive and migratory ability of HCC cells. Therefore, we speculate that eEF1A1 affects the invasion and metastasis of HCC by regulating HOXB9 expression. To confirm this hypothesis and explore its mechanism, RNA interference and other techniques are used to clarify the effect of eEF1A1 on the invasion and metastasis of HCC in vitro and in vivo by regulating HOXB9 expression. Then, luciferase reporter gene assay and chromatin immunoprecipitation (ChIP) are used to explore the specific mechanism of eEF1A1 regulating HOXB9. Finally, by detecting the expression of eEF1A1 and HOXB9 in HCC tissues, the correlation between the expression of eEF1A1 and HOXB9 and their significance as markers of post-operative metastasis and prognosis of HCC are analyzed. The results of this study will provide a new theoretical basis for elucidating the mechanism of invasion and metastasis in HCC.
