英文摘要
The methylation at N6 position of adenosine (m6A) is the most abundant RNA modification within protein-coding and long noncoding RNAs in higher eukaryotes. m6A is also the first RNA modification discovered to regulate gene expression and participate in a variety of critical biological processes. YTHDF proteins are the readers of m6A, the binding of which results in the alteration of the translation efficiency or stability of m6A-containing RNAs. Our published work demonstrate that YTHDF2 directly recruits the CCR4-NOT deadenylase complex to promote the deadenylation and decay of m6A-containing RNAs. However, the selectivity and regulation of m6A-mediated RNA decay still remain elusive. To further investigate the mechanism, we searched for novel regulators of CCR4-NOT complex by Mass-spectrum and found RNF219. In this study, we will first characterize the interaction between RNF219 and CCR4-NOT, and then take advantage of a well-established RNA decay monitor system to investigate the effect and mechanism of RNF219 in m6A-mediated RNA decay. In addition, we will identify the target genes of RNF219 in embryonic stem cells by RNA sequencing analysis and explore the function of RNF219 in early development.
RNA的稳定性是基因表达调控的重要机制之一。RNA的脱腺苷酸化,即poly(A)尾巴的去除,是细胞内大多数RNA降解的起始与限速步骤。在本研究中我们独立发现了脱腺苷酸化酶复合体CCR4-NOT的一个新的相互作用因子RNF219。在细胞内RNF219通过其C末端500-600aa与CCR4-NOT形成一个稳定的复合体。在小鼠ES细胞中敲低Rnf219导致Zscan4家族,转座子MERVL等二细胞特异转录本的显著下调。有趣的是,这些二细胞特异转录本在Cnot10敲低的小鼠ES细胞中发生了上调。我们还发现敲低Rnf219导致这类基因的poly(A)尾变短以及稳定性降低。进一步的报告基因脱尾实验证实了过表达RNF219能够抑制miRNA以及m6A修饰介导的RNA脱尾。RNF219的抑制作用依赖于其与CCR4-NOT复合体之间的相互作用而并不依赖于其RING finger结构域。而在RA诱导后,RNF219敲除的小鼠ES细胞中一些神经分化相关基因表达量低,并且不能像野生型一样诱导出神经元样细胞。于是我们认为RNF219是CCR4-NOT脱腺苷酸化酶的负调控因子,能够阻碍RNA的靶向性快速脱尾降解,从而维持ES细胞中的二细胞特异性转录本以及分化状态下的神经相关基因的正确表达,促进RA诱导的神经分化。
