大气细颗粒物PM2.5污染与脱髓鞘疾病的发展密切相关,但致病机制不清。我们前期研究发现小胶质细胞激活在PM2.5促髓鞘损伤中起关键作用,LBP9是介导PM2.5效应的特异表达基因,但其在小胶质细胞活化中的功能及信号转导机制未知。由于PM2.5的作用依赖于NFκB,LBP9启动子有保守的NFκB位点,我们提出PM2.5通过NFκB/LBP9信号轴诱导小胶质细胞激活加剧髓鞘损伤的机制假说。本课题将借助细胞特异性遗传操作和定向移植技术,明确PM2.5促髓鞘损伤的细胞基础(小胶质细胞)和特异效应基因(LBP9);进而通过ChIP-PCR、功能阻断/恢复实验,解析LBP9作为NFκB靶基因参与PM2.5诱导小胶质细胞激活的分子机制;最后利用脱髓鞘及髓鞘生成动物模型,系统评估靶向干预LBP9对PM2.5促髓鞘脱失的治疗作用。研究结果可为阐明PM2.5加剧脱髓鞘损伤的致病机制、筛选治疗靶点提供理论依据。
英文摘要
Atmospheric fine particulate matter (PM2.5) plays a critical role in the development of demyelinating diseases. However, the pathogenic mechanism remains unclear. In the previous study, we found that microglia activation is an important cause of PM2.5-induced exacerbation of myelin injury. LBP9 is a specifically expressed gene that responses to the PM2.5 stimulation, but its function and signal transduction in microglia activation is unknown. Given the impacts of PM2.5 is dependent on NFκB activation, and LBP9 promoter contains conserved NFκB binding domain, we thus hypothesize that PM2.5 induces microglial activation to aggravate CNS demyelination through NFκB/LBP9 signaling axis. In this proposal, we will firstly clarify the cytological basis (microglia) and specific responser (LBP9) of PM2.5 on exacerbating CNS demyelination, based on the cell-specific genetic manipulation and stereotaxic transplantation techniques. Then we will further elucidate the molecular mechanism of LBP9, as the NFκB target gene, involved in PM2.5-induced microglia activation, using CHIP-PCR and functional blocking/rescue assays. From the therapeutic angle, we will test the effects of LBP9 conditional deletion in microglia for the relief of PM2.5-induced demyelinating injury, based on using three demyelination/myelinogenesis animal models. This project, if successful, will elucidate the pathogenic mechanism of PM2.5-aggravated demyelinating diseases, and provide a new therapeutic target for the prevention and treatment of such diseases.
大气污染是我国面临的重要社会问题,阐明大气污染危害健康的作用机制是制定有针对性的防治策略和研发有效防治手段的关键。大气污染物中可吸入式悬浮细颗粒物(Particulate Matter,PM)是影响空气质量的关键指标,也是引起大气污染健康效应的重要物质基础。尽管越来越多的报道证实PM暴露会影响大脑发育,但PM与中枢神经系统疾病的相关性结论主要来自于流行病学研究,缺乏直接的实验证据和作用机制的探讨。本项目聚焦PM加剧中枢神经系统脱髓鞘性损伤的致病作用及分子机制这一国际前沿科学问题,首先通过炎性脱髓鞘模型研究了PM对炎症诱导的中枢髓鞘损伤及修复的影响,发现PM加剧髓鞘损伤,并激活外周免疫细胞和小胶质细胞。在Cuprizone诱导的毒性脱髓鞘模型中,发现PM加剧脱髓鞘并阻止再髓鞘化,以上两个互补的实验证实PM能通过血脑屏障加剧啮齿动物的髓鞘损伤并抑制髓鞘再生。另外,我们还发现孕期和围产期PM暴露抑制子代小鼠髓鞘生成,并增加中枢活化的小胶质细胞数量。离体实验显示PM刺激小胶质细胞的活性与LPS相似,可以诱导小胶质细胞向炎性状态转变。通过RNA-seq和ChIP-seq分析,揭示了PM通过TLR-4/NF-κB信号通路激活小胶质细胞,加剧神经炎症和髓鞘损伤。最后,通过探究中药蒲地蓝口服液和鞣花酸对PM诱导的髓鞘脱失的免疫调节作用,发现它们能有效抑制神经炎症,为预防和治疗相关疾病提供了理论依据。
