苎麻根系再生受阻导致的麻蔸代谢衰退,严重影响其地上部分的产量与品质。围绕解析苎麻根再生机制,推动再生能力强的品种选育这一科学问题与实际需求,采用植物发育生物学、生物信息学以及分子生物学技术与方法,以在植物根顶端分生区发挥重要作用的WUSCHEL-related homeobox(WOX)基因及其启动子区关键顺式作用元件为研究对象,重点研究:(1)不同苎麻种质BnWOX8启动子区生长素响应元件(AuxREs)重复序列多态性;(2)生长素响应元件多态性与BnWOX8表达水平的关联特征,以及BnWOX8对内源生长素应答机制;(3)筛选、鉴定BnWOX8调控的靶基因,绘制苎麻根系细胞命运转变阶段的分子和细胞学框架图谱。预期研究结果将阐明BnWOX8及其启动子区生长素响应元件对苎麻根系发育与再生的调控特征,加深对多年生宿根型作物根系发育与再生机制的理解,并为选育再生力强的苎麻新品种(系)提供参考。
英文摘要
The regeneration of the underground part of ramie was mainly through the de novo organogenesis of adventitious roots. While, most studies about ramie root regeneration were focused on the agronomic characteristic analyses. The transcription factor WUSCHEL-related homeobox are essential in the many stages of plant development by promoting cell proliferation. Our previous analysis showed that the promoter region of ramie WOX contain several auxin response elements (AuxREs), and the copy number polymorphism of AuxREs in BnWOX8 from different ramie resources were observed. It was presumed that this BnWOX8 gene may play vital role in de novo adventitious root organogenesis of ramie. In this study, we try to elucidated the regulatory mechanism underling the cell fate transition, which links the upstream wounding and hormonal signaling to the downstream organ formation using our simple de novo root organogenesis system that mimics the natural conditions without added hormones. The result of the study will help to better understand the de novo organogenesis mechanism of perennial crops and provide new insights for rapid ramie propagation and breeding.
