核酸富集技术可以特异性的从混合基因样品中富集及回收目标核酸,并进一步通过测序等方法鉴定目标核酸。应用核酸富集技术既可以从环境微生物、宏基因组中发掘低拷贝的新型天然产物,也可以进行相关疾病的基因检测和基因诊断。因此,开发一种快速、稳定、高特异性的核酸富集新技术具有非常重要的理论和现实意义。本研究拟将Cas9-BE3碱基编辑系统与尿嘧啶结合蛋白UdgX有机结合,通过设计不同的靶标sgRNA,实现对目标核酸的特异性标记、富集及回收。这种基于CRISPR-Cas系统的新型核酸富集技术可应用于基因检测、天然产物挖掘等领域,填补相关研究的空白,为其它基因检测技术提供新思路,增强未来生物技术高端产品研发的核心竞争力,具有较大的实践价值。
英文摘要
Nucleic acid enrichment technology can specifically enrich and recover the target nucleic acid from the mixed gene samples, and further identify the target nucleic acid by sequencing and other methods. The application of nucleic acid enrichment technology can not only discover new natural products with low copy from environmental microorganisms and macro genomes, but also carry out gene detection and gene diagnosis of related diseases. Therefore, it is of great theoretical and practical significance to develop a rapid, stable and highly specific technology for nucleic acid enrichment. In this study, Cas9-BE3 base editing system is combined with UdgX, and different target sgRNA is designed to realize the specific labeling, enrichment and recovery of target nucleic acid. This new nucleic acid enrichment technology based on CRISPR-Cas system can be applied to gene detection, natural product mining and other fields, fill in the blank of related research, provide new ideas for other gene detection technologies, and enhance the core competitiveness of high-end product research and development of biotechnology in the future, which has great practical value.
