Signal communication between nucleus and chloroplasts are necessary for biogenesis of chloroplasts, synergetic expression between nucleus genome and chloroplast genomes, as well as regulation to photosynthesis. VAR6/PAC is one of the essential chloroplast localized factors, encoded by nucleus genome, to regulate expression of chloroplast genomes. var6-1 is the VAR6/PAC knock-down mutant with albino ture leaves at early stage, and with decreased accumulation of photosynthetic subunits. Suppressor mutants of var6-1 were screened defined by green true leaves and also recovery accumulation of photosynthetic subunits. The locus of S04-03 suppressor and the S05-31A suppressor was determined as Nup160 and Nup96 respectively, by map based cloning and complementation assay. Both Nup160 and Nup96 are nucleoporins of the Nup107-Nup160 subcomplex belonging to nuclear pore complex (NPC). These are genetic evidences that NPC regulates signal communication between nucleus and chloroplasts. In this research, we are going to discover the accumulation of signal molecules in the nucleus of NPC mutants with cell biology combined transcriptome and proteome strategies, to finally establish the model of signal communication between nucleus and chloroplasts regulated by NPC.
