甘遂与甘草配伍属于中药配伍禁忌“十八反”的范畴,但二者能否同用一直存在争议。根据中医药证相关理论,二者能否同用与机体状态不同有关。本课题组前期的研究表明,癌性腹水模型下,甘遂半夏汤中甘遂甘草反药组合在利水方面属“增效”的配伍关系,产生“增效”的机制是什么?本课题组前期研究发现含甘遂甘草反药组合的甘遂半夏汤具有免疫调节和排钠利尿作用。因此本课题结合前期的研究结果设想甘遂甘草反药组合在利水方面发挥“增效”的机制可能与甘遂甘草配伍后抑制TLR4-MyD88/NF-κB通路的激活,发挥抗肿瘤、免疫调节而减少腹水生成有关,也可能与二者配伍后促进NPs-NPRs-cGMP-PKG通路的激活而发挥排钠利尿,增加水饮排出有关。因此通过检测上述两条通路相关指标,从抑制腹水生成和促进水饮排出两个方面探究甘遂甘草反药组合在利水方面发挥“增效”的机制,以期验证中药及复方治疗疾病具有多通路、多靶点、综合调节的特点。
英文摘要
Gansui and Gancao compatibility belongs to the category of “Eighteen antagonisms” which is an part of the controversial prohibited combinations in traditional Chinese medicine. According to the theory of TCM, whether Gansui and Gancao can be put together is associated with disease model. The results of the preliminary research showed that under the model of cancerous ascites, Gansui and Gancao anti-drug combination play a role of "synergistic effect" in Gansui Banxia Decoction. So what is the mechanism for the "synergistic effect "? The results of our preliminary research showed that Gansui Banxia Decoction with Gansui and Gancao anti-drug combination has the functions of immune regulation and sodium excretion and diuresis. so this study combined with previous research results envisage that the mechanism of the synergistic effect of Gansui and Gancao anti-drug combination on alleviating water retention may be related to Gansui and Gancao anti-drug combination can inhibit activation of TLR4 - MyD88 / the nf-kappa B pathway, play anti-tumor, immune regulation and ultimately achieve formation of reducing ascites, also may be related to Gansui and Gancao anti-drug combination can promote activation of NPs - the NPRs - cGMP - PKG pathway and play a function of diuretic and ultimately increase water discharge. Therefore, we will explore the mechanism of the synergistic effect of Gansui and Gancao anti-drug combination on alleviating water retention from reducing ascites and increasing water discharge by detecting the related indexes of the above two pathways, so as to verify the Chinese medicine which has the characteristics of multi-pathway, multi-target and comprehensive regulation when they will be used to treat diseases.
