神经递质多巴胺受体基因DRD2是抗精神病药物的主要靶标,但临床有效的抗精神病药物均可阻断D2多巴胺受体引发严重且威胁生命的副作用,已有动物模型不能满足DRD2基因精神疾病病理机制和药物研究。家犬中大脑神经递质相关基因与人类存在趋同进化,具有和人类相似的脑结构、行为表型和药理反应,是DRD2基因研究的极佳材料。本项目将依托已获得F0代DRD2基因编辑犬杂合子,在利用遗传学和生理学研究手段明确F1代DRD2基因编辑犬遗传背景和健康状态的基础上,基于申请人及研究团队行为学和脑影像学研究基础,对F1代DRD2基因编辑犬行为及脑区结构和功能进行研究,阐明该编辑犬行为与精神疾病行为表型相似性,揭示影响该编辑犬行为的脑区结构与功能变化。研究结果为后续理解DRD2基因相关精神疾病机理以及开发新疗法和临床前药物测试等应用奠定基础,拓展家犬作为动物模型在精神疾病相关研究的应用。

The neurotransmitter dopamine receptor gene DRD2 is the main target of antipsychotic drugs, but clinically effective antipsychotic drugs can block the D2 dopamine receptor and cause serious and life-threatening side effects. The existing animal models cannot satisfy the requirements of the pathological mechanism and drug research of psychiatric diseases with DRD2. The brain neurotransmitter-related genes in domestic dogs have convergent evolution with humans, and they have similar brain structures, behavioral phenotypes, and pharmacological responses with those in humans. Therefore, they are excellent materials for the study of DRD2 gene. This project will rely on the obtained F0 generation DRD2 gene-edited dog heterozygotes, use genetic and physiological research methods to clarify the genetic background and health status of the F1 generation DRD2 gene-edited dog, and use existing behavioral and brain imaging research foundation of the applicant and the research team, to study the behavior and brain structure and function of F1 generation DRD2 gene-edited dogs, to clarify the similarity between the behavior of the edited dog and the behavioral phenotype of psychiatric disorders, and to reveal the changes in the structure and function of the brain area that affect the behavior of the edited dog . The research results lay the foundation for the follow-up understanding of the mechanism of DRD2 gene-related psychiatric disorders, the development of new therapies and preclinical drug testing and other applications, and the expansion of the application of domestic dogs as animal models in psychiatric disorders -related research.
本研究围绕神经递质多巴胺受体基因DRD2这一抗精神病药物的重要作用靶点展开。针对现有动物模型难以满足DRD2基因功能研究和药物研发需求的现状,利用家犬与人类在神经递质系统上的高度保守性,以及其在脑组织结构、行为表型和药理反应等方面与人类的相似性,创新性地构建了DRD2基因编辑犬模型。并利用获得的DRD2基因编辑杂合子犬,系统开展了编辑犬的生长发育监测、生理生化指标检测、行为学评估以及脑结构与功能分析。主要研究结果包括:(1)基因编辑效率达到47.3%,全基因组测序证实编辑特异性良好,未发现脱靶效应;(2)编辑犬生长发育指标正常,未出现明显生理异常;(3)行为学分析显示编辑犬存在显著的社交,尤其是跨物种社交障碍;(4)神经影像学研究发现编辑犬在尾状核、嗅球、扣带回等关键脑区出现明显的结构和功能改变。这些发现为阐明DRD2基因在神经精神疾病中的分子机制提供了重要的实验证据。基于本研究的创新性发现,我们将进一步深入解析DRD2基因调控神经精神疾病表型的时空特异性,阐明关键神经环路的分子机制,为精神疾病的发病机制研究和创新药物研发提供新的理论依据和实验平台。本研究的顺利完成为后续获得更高级别的项目资助奠定了坚实基础。
