Abstract An environmental stress,such as drought,high-salt and cold stresses,has become a primary restriction factor. So the genetic improvement of plant resistibility using modern biotechnology is an universal task in the fields of molecular biology. Recent years,some response genes to environment stresses have been cloned from a variety of plants,which functions are all relative to plant transcription factor.The transcriptor factors containning AP2/EREBP domain form a large gene family, all the proteins of the AP2/EREBP family contain one(or two)DNA-binding domain(AP2/EREBP domain)composed of 57-70 amino acid residues,a nuclear localization signal and a putative transcription activation domain.All of them play an important role in developmental processes,signal conduction and resistibility.It is helpful for elucidating the mechanism of stress signal conduction in plant cells to study the interaction between transcriptor and cis-acting element.. Using the yeast one-hybride analysis system ,we demonstrated that TINY could function as a DREB-like transcription factor specifically binding to the DRE cic-acting element.We isolated one maize DREB protein ,named maDREB1,which function as DREB-like transcription factor too. Point mutation analysis showed that V14 and E19 were both important in protein-DNA interacting in maDREB1,though V14 was more essential. .
