炎症性肠病(IBD)是一组原因不明的慢性肠道炎症性疾病,影响生活质量。ROS水平显著增加导致的机体氧化应激和炎症反应增强使得肠上皮屏障功能损伤是IBD发生和发展的关键病理环节。氧化应激可以通过NF-κB信号通路促进NLRP3的聚集及活化,导致IBD的发病和进展。红萸饮(HYY)是治疗IBD的验方,临床疗效显著,其组分能显著抑制ROS的产生。课题组前期研究发现,HYY可缓解IBD小鼠结直肠炎症;其组分也能通过抑制NF-κB信号通路缓解肠上皮细胞炎症反应,但具体机制不明。本项目拟在前期研究的基础上,从体内、体外两个水平开展工作,探究HYY保护IBD肠上皮屏障功能的潜在机理,证明其可以通过抑制氧化应激,调控ROS/NF-κB/NLRP3信号通路,抑制NLRP3的表达,减少炎症反应,保护肠上皮屏障功能,缓解IBD,为HYY治疗IBD提供理论基础与实验依据。

Inflammatory bowel disease(IBD) is a group of chronic intestinal inflammatory diseases with unknown causes, which greatly affects the quality of life. The significant increase of ROS level leads to the increase of oxidative stress and inflammatory response in the body, which makes intestinal epithelial barrier function damage becoming a key pathological link in the occurrence and development of IBD. Oxidative stress can promote the aggregation and activation of NLRP3 through NF-κB signaling pathway, leading to the occurrence and progression of IBD. Hongyuyin Decoction(HYY) is a proved recipe for the treatment of IBD, with significant clinical effect, and its components can significantly inhibit the production of ROS. Our previous studies found that HYY can alleviate colitis in IBD mice. The components of HYY can also alleviate the inflammatory response of intestinal epithelial cells by inhibiting NF-κB signal pathway, but the specific mechanism is unknown. On the basis of previous research, this project intends to work on two levels, in vivo and in vitro, to explore the potential mechanism of HYY to protect the intestinal epithelial barrier function of IBD. This project also intends to prove that HYY can inhibit oxidative stress, regulate ROS/NF-κB/NLRP3 signaling pathway, inhibit NLRP3 expression, reduce inflammatory response, protect intestinal epithelial barrier function, and relieve IBD, in order to providing the theoretical basis and experimental basis for HYY treatment of IBD.
