蜜腺是一类能为传粉者提供报酬的重要结构,在植物和传粉者的互作中扮演了极其重要的角色。因此,一直以来,蜜腺都是生态-进化-发育生物学领域的研究热点。前人关于蜜腺形成机制的研究主要集中在真双子叶植物,尤其是核心真双子叶植物。由于缺乏合适的研究体系,单子叶植物中蜜腺形成的调控机制尚不清楚。姜科(Zingiberaceae)象牙参属(Roscoea)植物不仅具有其它单子叶植物不具有的位于子房顶端的上位蜜腺,并且具有发育正常且能分泌蜜汁和发育不正常不能分泌蜜汁的两种类型的蜜腺,这使得其成为了开展蜜腺发育分子机制研究的理想体系。本项目拟在已有工作的基础上,结合多种技术手段,追踪不同类型蜜腺的发育过程,揭示不同类型蜜腺的差异在发育过程中是如何建立的;解析不同类型蜜腺发育的分子机制并推测象牙参属植物中蜜腺演化产生的过程。本项目的开展将为我们理解单子叶植物蜜腺的发育和演化提供新的见解。
英文摘要
Nectary, which is an important structure that can provide rewards to pollinators, plays an essential role in the interaction between plants and their pollinators. Therefore, nectary has been being one of key issues in evolutionary developmental (Evo-Devo) biology. Previous studies concerning about the mechanisms underlying nectary development are mainly focused on eudicots, especially core-eudicots. However, due to the lack of suitable systems, the mechanisms underlying nectary development in monocots remain unknown. The genus Roscoea in Zingiberaceae is an exceptionally ideal group for studying the mechanisms underlying nectary development, because it has special epigynous nectaries on the top of carpel, which do not occur in other monocots. Meanwhile, two types of nectaries can be categorized based on the structure and the ability to secrete nectar: one is the nectary which develops normally and secretes nectar, and the other is the nectary which develops abnormally and does not secrete nectar. In this project, on the basis of previous studies and by combining various multidisciplinary technologies, we aim to: trace the developmental processes of different types of Roscoea nectaries and find out when the differences are established; elucidate the mechanisms underlying the development of nectaries in Roscoea and then propose the evolutionary process of nectary in Roscoea. This project will provide insights into the development and evolution of nectaries in monocots.
蜜腺是能分泌蜜汁的腺体结构,其发育和进化机制一直是经典研究议题。目前关于蜜腺发育机制的研究主要聚焦于核心真双子叶植物和部分基部真双子叶植物,但是,单子叶和其他类群植物蜜腺发育的机制仍不清楚。在本项目中,我们以单子叶植物类群之一的姜科象牙参属中分别具有发育正常且能分泌蜜汁和发育不正常不能分泌蜜汁的两种类型蜜腺的两个物种(早花象牙参和无柄象牙参)为研究对象,通过结合多种方法和技术揭示了该两个物种蜜腺在外观形态和内部解剖结构、蜜汁分泌动态、蜜汁成分的动态变化与传粉者的关联、蜜汁分泌的关键基因以及不分泌蜜汁的原因。基于形态学和解剖学的研究,我们发现早花象牙参与无柄象牙参的蜜腺在大小、颜色及形状等方面存在着显著差异;在整个开花期,早花象牙参均能分泌花蜜,而无柄象牙参则不能;早花象牙参蜜腺分泌表皮细胞的细胞质浓厚并伴随淀粉粒的沉积,而无柄象牙参的分泌表皮细胞的细胞质浓度较低且未见淀粉粒沉积。传粉生物学研究发现,早花象牙参的有效传粉者熊蜂的访花频率与花蜜分泌的动态变化相契合。代谢组测定和分析发现,早花象牙参花蜜的代谢物种类丰富,涵盖了多种化学结构和功能类型的分子,且其中除了含量较为稳定的糖类物质外,还含有多种非糖类物质,非糖类物质在一天内不同时段存在着显著的变化。通过联合转录组和代谢组,我们发现了早花象牙参花蜜成分的相关调控网络。更为重要的是,通过比较两个物种蜜腺中表达的基因,我们发现了调控早花象牙参蜜汁分泌的关键基因Rc026561,该基因为SWEET9基因的同源基因。而无柄象牙参中该基因的同源基因Rs022800在3’端发生了缺失,可能导致其不能分泌蜜汁。在喙花姜中,其同源基因中的一个拷贝也在蜜腺中特异高表达,因此,我们推测SWEET9基因对于蜜汁分泌的作用可能在单子叶植物中就已经出现,而不是仅在核心真双子叶植物中被招募。本研究为单子叶植物蜜腺发育调控网络的揭示奠定了基础,同时为整个被子植物中蜜腺演化机制的解析提供了新证据。
