能量代谢紊乱是肿瘤细胞区别于正常细胞的特征性改变。LKB1-AMPK激酶是细胞能量代谢的关键感应及调节激酶,在维持上皮细胞极性中也起着重要作用。临床资料证实LKB1基因在乳腺癌细胞常发生杂合性缺失,其表达与乳腺癌恶性度呈负相关,然而目前对LKB1-AMPK激酶通路在乳腺癌发生发展中的细节机制尚不清楚。我们的前期研究结果显示,LKB1下游激酶AMPK参与调控细胞纺锤体检查点蛋白BUB1及上皮细胞标志物E-Cadherin,从而与维持染色质稳定性及上皮细胞特性有关。因此,我们设想:LKB1-AMPK激酶异常可导致细胞染色质不稳定;同时引起细胞极性丢失及上皮间皮化并导致肿瘤侵袭转移。我们拟以乳腺癌为研究对象,对LKB1-AMPK激酶异常在乳腺癌发生发展中的作用进行研究。该项目的完成,将阐明LKB1-AMPK激酶异常在乳腺癌恶性进程中的全新作用机制,并为筛选抗肿瘤药物的新靶点提供理论依据。
英文摘要
Deregulation of cellular energy metabolism is one of the cancer hallmarks. LKB1-AMPK kinase pathway plays a pivotal role in sensing and regulating cell energy metabolism and in maintaining cell polarity. Clinical data has shown loss of heterozygosity of LKB1 in breast cancer and negative correlation of LKB1 expression with the histological grade of breast cancer. However, the mechanism underlying the role of LKB1-AMPK kinase pathway dysfunction in the malignant progression of breast cancer is still unclear. Our preliminary data have shown that LKB1-AMPK kinase pathway may play an important role in regulating spindle checkpoint protein BUB1 and epithelial marker E-cadherin, which may indicate that LKB1-AMPK kinase pathway may be related to the maintenance of chromosome integrity and epithelial characteristics. Based on our preliminary data and previous literature reports, we hypothesize that dysfunction of LKB1-AMPK kinase pathway may lead to cell chromosome instability and epithelial-messenchymal transition (EMT), and consequently, give rise to breast cancer metastasis. Based on our preliminary data, we plan to conduct research on the effects of deregulated LKB1-AMPK kinase pathway on the malignant progression in human breast cancer, and to clarify a novel mechanism by which LKB1-AMPK kinase pathway affects the malignant progression of breast cancer.This project is expected to provide a theoretical basis for screening a novel anti-tumor target.
乳腺癌是女性最常见的恶性肿瘤之一,具有发病率高、起源复杂、组织结构多样、受激素调节等特点,长期以来一直是最受关注的女性肿瘤之一。能量代谢紊乱是肿瘤细胞区别于正常细胞的重要特征性改变。LKB1-AMPK激酶既是细胞能量代谢的关键感应及调节激酶,又是调控细胞极性的重要激酶。所以本项目以LKB1-AMPK激酶系统为研究对象,探讨其于乳腺癌恶性进程的关系及具体调节机制。.本项目通过改变LKB1-AMPK激酶表达水平研究其对乳腺癌细胞极性、侵袭转移能力、染色质稳定性、血管生成等方面的影响。利用免疫印迹、免疫荧光、免疫共沉淀、三维培养、TALEN技术等研究手段阐明LKB1-AMPK激酶发挥功能的分子机制;在移植瘤模型中阐明了AMPK激动剂二甲双胍对乳腺癌血管生成的调节作用,为乳腺癌的综合防治提供了新的方向。.本项目的研究结果证实LKB1表达与乳腺癌恶性程度成负相关,与ER/PR水平正相关。LKB1-AMPK激酶通路失活引起乳腺癌细胞发生EMT,同时极性紊乱及侵袭转移增加。 LKB1-AMPK激酶通路中AMPKα1与细胞纺锤体检查点蛋白BUB1相互结合,维持染色体稳定性。LKB1-AMPK激酶通路的下游极性蛋白CRB3可通过调节细胞极性、增值侵袭及干细胞特性影响乳腺癌恶性进程。在腺癌4T1细胞移植瘤模型中证明AMPK激活剂二甲双胍在乳腺癌肿瘤血管生成中的作用机制。.本项目的研究内容丰富了现有对LKB1-AMPK激酶系统的认识,补充了目前对于LKB1-AMPK激酶系统调节的信号网络的认识。本项目从细胞极性、染色质稳定和血管生成等方面,证实了LKB1-AMPK通路在乳腺癌恶性进程的重要调节作用,,为筛选抗肿瘤药物的新靶点提供理论依据。
