Hepatocellular carcinoma (HCC) is the main form of primary liver cancers, and is one of the main causes of cancer-related death in China. Epithelial-mesenchymal transition (EMT) plays an important role in promoting cancer cell motility and malignant development in HCC. TGF-β is one of the major inducers of EMT and HCC cell motility in vivo, and the spatio-temporal specific signaling regulation is crucial for TGF-β to exert its functions in this regard. KLF2 is a member of the KLF transcription factor protein family. Our unpublished data indicate that KLF2 could inhibit TGF-β/Smad signaling through a feedback circuit in HCC, attenuating TGF-β-induced expression of EMT-related genes and cancer cell motility, such as migration and invasion. This proposal aims to elucidate the molecular mechanisms underlying KLF2-mediated feedback regulation of TGF-β signaling and their roles in HCC cell EMT and motility. The expected results would further our understanding as to how TGF-β signaling is regulated by KLF2, and how this regulation makes sense in liver cancer cell EMT and motility. Furthermore, the study may also provoke new strategies or provide new molecular targets for targeted intervention in liver cancer.
肿瘤细胞的可塑性参与了肿瘤细胞异质性的构筑,而肿瘤细胞异质性又是肿瘤侵袭转移和耐药的基础。理解肝癌细胞命运转变及其可塑性的分子调控机制不仅有助于揭示肿瘤发展的原因,而且有可能为肿瘤诊断和分子靶向干预提供新策略、新靶点和理论依据。本项目重点探讨细胞内主要信号通路异常以及关键蛋白质在肿瘤发展中的功能与机制。本项目研究发现:KLF2在肝癌中低表达与病人不良预后密切相关;TGF-β能在肝癌细胞系中刺激KLF2基因表达;过量表达KLF2能够在肝癌细胞中抑制TGF-β/Smad信号转导,从而形成一个新的负反馈调控环。进一步机制研究表明,KLF2通过抑制Smad蛋白的转录调控活性而减弱TGF-β诱导的靶基因表达,包括EMT相关基因等,从而减弱TGF-β诱导的肝癌细胞运动。此外,本项目还深入研究了TGF-β信号和Hippo通路协同调控CYR61基因表达的作用与机制。项目成果揭示KLF2拮抗TGF-β信号抑制肝癌发展的新功能,探究了TGF-β信号和Hippo信号对话的新机制,为肿瘤干预提供新思路和新靶点。本项目总计发表高水平SCI论文2篇,包括J Biol Chem(2024)、Front. Immunol(2024),培养博士研究生1名和硕士研究生2名。
