围绕我们新近发现并率先向GenBank数据库注册、并已申请中国发明专利的新型人源神经分蜃樱╤NDF)进行研究,拟采用Northern Blot、原位杂交技术等研究hNDF基因在大鼠神经系统发育过程中的表达图谱;构建表达载体,进而探讨hNDF基因与神经细胞分化(以神经母细胞瘤为主)的关系,探讨hNDF作为一种新的肿瘤分化诱导剂应用于临床的可能。

The project was aimed to study the function of a novel gene named "nerve differentiation factor (hNDF for abbreviation)" which was discovered in our lab. Full-length cDNA sequence of hNDF is 1990bp and capable of encoding a novel protein of 187 amino acids. The similarity of hNDF is 50% compared with the known mouse derived nerve differentiation factor TA20 in amino acid sequence level. A set of experiments were used to test the biological activity of hNDF. Results demonstrated that hNDF has similar biological function with its homology member TA20 in stimulating the differentiation of NG108-15 cells and the growth of ganglion cells. Tissue expression analysis using multiple tissue dot blot also demonstrated that hNDF was also extensively expressed in many tissues other than the nervous system, which suggest much more functions of hNDF in the body.
