氨基酸的补充一直是人们所关注的问题,小肽(二肽和三肽)可以短时间内快速大量补充氨基酸,但是小肽(二肽和三肽)结构和氨基酸补充速率之间的关系尚不明确。因此本项目以小肽(二肽和三肽)为研究对象,重点研究PepT1载体参与的氨基酸补充过程。通过构建基于PepT1转运途径的小肽补充氨基酸的细胞模型,建立并优化氨基酸补充速率的测定方法;利用氨基酸描述符等参数化肽结构,结合神经网络等方法,探明小肽(二肽和三肽)结构与氨基酸吸收速度之间的关系;通过理论计算阐明小肽(二肽和三肽)与PepT1之间的分子结合模式。最终明确基于PepT1载体转运途径的小肽(二肽和三肽)结构和氨基酸补充速率之间的关系及机理。从而进一步完善氨基酸补充途径和机理,同时为快速补充氨基酸的小肽筛选、加工和产品开发提供新的理论依据,为食源性小肽的研究提供新的思路。

Amino acid supplementation is a key question concerned by people. Small peptides(dipeptide and tripeptide) can quickly supplement a large amount of amino acids in a short time. However, the relationship between the structure of small peptides (dipeptide and tripeptide) and the rate of amino acid supplementation is still unclear. Therefore, we take small peptides (dipeptide and tripeptide) as the starting point, focus on the amino acid supplement process involved in the PepT1 carrier. By building a cellular model based on the PepT1 transport pathway, we will establish and the method for determining the rate of small peptide (dipeptide and tripeptide) supplementing amino acids. With different amino acid descriptors and other parameters, we will establish a quantitative structure-activity relationship between the structure of small peptides (dipeptide and tripeptide) and the ratio of supplementing amino acids. We will also investigate the binding mode between the small peptides (dipeptide and tripeptide) and PepT1. This project will provide a new theoretical basis for the screening processing and developing of small peptides. This project can also provide new ideas for the research of food-derived small peptides.
本研究构建了基于PepT1载体转运途径的小肽补充氨基酸模型,重点研究了小肽的吸收速率、构效关系及其在泌乳功能中的作用。首先,采用转染HEK293T细胞建立了小肽吸收模型,经过优化表达载体、细胞体系和表达条件,成功构建了基于HEK293T的PepT1载体转运模型。通过RT-qPCR、Western Blot及荧光蛋白标记技术确认了PepT1载体的过表达,验证了该模型的可行性。为了进一步验证实验结果,研究还建立了基于ICR小鼠的氨基酸快速补充模型,开展了不同小肽吸收速率的研究,并建立了小肽补充氨基酸速率的数据库。在构效关系分析方面,研究结合分子动力学模拟,建立了二肽转运自由能的评价体系,并通过增强采样模拟分析了PepT1底物的识别机制。结果表明,二肽的两端电荷分布对底物结合具有重要影响,静电相互作用促进了底物的结合。进一步的自由能计算发现,带负电侧链的二肽不利于结合。通过对二肽进行侧链、C端和N端的屏蔽,验证了已构建模型的可靠性,同时通过PepT1突变实验进一步验证了该模型的准确性。研究结果揭示了电荷、疏水性、范德华体积及氢键等结构特征在小肽结合稳定性和转运能力中的关键作用。基于该模型,研究合成了促进泌乳功能的蛋氨酸二肽(EM),并进行了相关分析。结果表明,EM小肽能够有效促进泌乳。此外,还利用预测结果指导制备了蛋清蛋白肽,实验表明该肽也具有显著的泌乳促进效果,并且能够促进子鼠的生长发育及维持其肠道菌群的平衡。本研究申请了1项国家发明专利,并发表了2篇SCI检索论文,培养和协助培养了2名硕士研究生。总之,本研究为小肽的转运机制、构效关系分析及其在泌乳促进中的应用提供了重要的理论基础和实验数据。
