结肠炎慢性炎症持续发展可引起结肠炎相关性癌变（CAC）。晚近，发现环状RNA circ-ITCH在结直肠癌中表达异常下调，circ-ITCH可以通过发挥miRNA海绵作用抑制Wnt/β-catenin信号通路，抑制肿瘤细胞增殖，可能是circ-ITCH参与CAC发生发展的关键环节。本课题在艾灸治疗UC有效以及化脓灸能提高消化道肿瘤患者免疫功能的基础上，拟以circ-ITCH为切入点，从动物实验、临床研究结合细胞干扰实验来求证：艾灸可能是通过影响circ-ITCH与miR-7/miR-214的分子海绵作用增强ITCH的表达，促进磷酸化形式的蓬乱蛋白2泛素化降解从而达到抑制Wnt/β-catenin信号通路的作用，以达到艾灸抑制肠道炎症、阻遏结肠炎相关性癌变之目的，初步阐明艾灸对CAC发病的癌变关键circRNA的作用机制，以期为艾灸在结肠炎及其癌变中的临床应用提供实验资料和科学依据。

The continuous development of chronic inflammation of colitis can cause colitis-associated colorectal cancers (CAC). Recently, it was found that the expression of circ-ITCH is down-regulated in colorectal cancer. Circ-ITCH can inhibit the Wnt/β-catenin signaling pathway by playing the role of miRNA sponge and inhibit the proliferation of tumor cells, which may involves in CAC pathogenesis. Moxibustion is effective for UC and can improve the immune function of patients with gastrointestinal tumors. The aim of this project is to use circ-ITCH as an entry point to verify from animal experiments and clinical studies combined with cell interference experiments: moxibustion may enhance the expression of ITCH by affecting the molecular sponge effect of circ-ITCH and miR-7/miR-214, and promote the ubiquitination degradation of phosphorylated DVL2 to inhibit Wnt/β-catenin signaling pathway. To confirm that moxibustion therapy has an effect on circRNA to inhibit colitis and its carcinogenesis, so as to provide an objective scientific basis for the clinical application of moxibustion in colitis and its carcinogenesis.
