昆虫对杀虫剂敏感性存在性别差异的现象普遍存在,但其内在的分子机制尚不清楚。申请人前期研究发现雄性灰飞虱对新型介离子类杀虫剂三氟苯嘧啶比雌性更加敏感,其中2个P450基因CYP314A1和CYP439A1v3能够被三氟苯嘧啶诱导上调且在雌虫中高表达。推测这2个基因的高表达导致灰飞虱雌虫对三氟苯嘧啶解毒代谢能力增强。据此,本项目拟1)通过RNAi沉默CYP314A1和CYP439A1v3基因,探明这2个基因在雌雄灰飞虱对三氟苯嘧啶敏感性差异中的作用;2)通过体外真核表达CYP314A1和CYP439A1v3蛋白并代谢三氟苯嘧啶,确定其在三氟苯嘧啶解毒代谢中的催化作用;3)通过荧光素酶检测系统确定CYP314A1和CYP439A1v3的调控元件和转录因子,明确其转录调控机理。研究结果将揭示灰飞虱对三氟苯嘧啶敏感性存在性别差异的分子机制,为灰飞虱的有效防治提供理论依据。
英文摘要
It is a common phenomenon that male and female insects always show different susceptibility to insecticides, while the mechanisms are not clear. Our previous studies showed that females are more resistant to triflumezopyrim than males in Laodelphax striatellus. In addition, the expression of CYP314A1 and CYP439A1v3 could be induced by triflumezopyrim and overexpressed in females of L.striatellus than that in males, which might enhance the ability of detoxification of triflumezopyrim. In this study, the functions of CYP314A1 and CYP439A1v3 in sex differences of susceptibility to triflumezopyrim in L. striatellus will be studied using RNAi. Then the both P450 genes will be expressed in vitro using the eukaryotic expression system, and determine their detoxification activity to triflumezopyrim. At last the regulation mechanism of CYP314A1 and CYP439A1v3 expression will be investigated at transcription level using the luciferase reporter assay. The results of this study will reveal the molecular mechanism of gender difference in the susceptibility of L.striatellus to triflumezopyrim, and provide theoretical basis for effective control of this pest.
