细胞重编程过程中存在两种细胞群体:成功进行重编程的细胞和不能进行重编程的细胞,我们的前期研究发现它们之间存在细胞通讯而且对于iPS的产生具有重要的作用,具体的机制还未知。本项目以诱导多能干细胞过程和多能干细胞始发态/原始态转变两个细胞重编程体系为模型,运用前沿的技术手段系统研究细胞群体之间的通讯对表观遗传修饰和细胞代谢的影响,进而参与细胞群体命运转变的调控,揭示细胞通讯对细胞命运转变的基本规律和机制。对体外细胞重编程过程中细胞通讯模式的探究将会促进体内组织和器官形成过程中细胞间相互作用的研究,并且为各种疾病的发生发展提供新的研究视角。
英文摘要
There are two kinds of cell population in the process of somatic reprogramming: reprogrammable and non-reprogrammable cells. Our preliminary research data suggests the communications between this two cell-population play important roles in the generation of iPS, but the detailed mechanisms are unknown. The proposed study will focus on exploring the essential rules and mechanisms of cell communications in two defined cell fate transition: acquired pluripotency of somatic reprogramming and prime-naïve pluripotency transition. We will utilize state-of-the-art molecular biology and biochemistry platform in our lab to investigate how cell communications between cell population change epigenetic modifications and metabolism in cell driving cell fate conversation. The study can provide valuable model to investigate cell communications involved in tissues and organs formation in vivo, furthermore the proposed project could inspire the research of different diseases including initiation and development of cancers with new insights.
本项目研究了2种体细胞重编程多能干细胞诱导体系和1种多能干细胞primed态向naïve态转变的诱导体系中细胞分群情况、细胞命运转变情况。发现2种体细胞重编程产生多能干细胞的诱导过程中,存在走向重编程失败和成功分支的细胞群;解析了不同命运分支细胞簇特异的调控子网络,进一步分析了重编程失败和成功细胞簇间的互作活跃的配体受体对。解析了诱导体系共有和特异的参与调控重编程成功和失败的细胞命运的下游关键信号通路。而由BMP4介导的多能干细胞primed态向naïve态转变的诱导过程中,产生了一类PGC样的细胞群体。发现了在多能干细胞primed态向naïve态转变的诱导体系中存在PGCLCs的中间态细胞群体,确定了PGCLCs是多能干细胞primed态向naïve态转变过程中必经的中间状态且中胚层基因T的正常表达对于PGCLCs的中间态细胞群体的产生和naïve态多能性的建立使必需的。且Zbtb7a/b基因可以调控PGCLCs的中间态细胞群体的产生。此外,鉴定出Gata2是placenta细胞分支重要的调控因子。本项目还分选并证明了BVSV双阳细胞具备分化成生殖细胞的能力。证明了小鼠中在多能干细胞primed态向naïve态转变的诱导体系中存在PGCLCs的中间态细胞群体,且PGCLCs是多能干细胞primed态向naïve态转变过程中必经的中间状态。从单细胞水平上揭示了小鼠始发态向原始态多能性转变的新型分子路径和调控机制,为研究人的胚胎发育研究提供了新的理论参考,同时也为PGCLCs特化为PGC提供了高效的体外研究模型,鉴定分离得到了能够具备生殖细胞分化能力的PGCLCs中间态细胞簇,也为人类生殖问题的研究提供了细胞模型。
