人类肠道微生物类群丰富,细菌数量可达100万亿个。随着分子生物学技术的不断发展,肠道微生物逐渐被发现与人类疾病息息相关,成为研究热点,但一些肠道致病菌的不可培养,限制了一些疾病致病机理的研究。获得纯培养成为了研究肠道细菌的互作关系、生理特性、致病机制的基础和瓶颈。本项目拟通过设计基于MPN前培养体系,添加藤黄微球菌复苏促进因子(Rpf)等的组合培养方法,对人体肠道的细菌进行可培养研究。同时,结合肠道微生物宏基因组数据,探索肠道微生物VBNC状态的形成和复活机制。本项目旨在提高肠道VBNC状态细菌的可培养性,同时为肠道致病菌的可培养提供理论基础和技术支撑。

Human intestinal microbial groups are abundant, the number of bacteria can reach 100 trillion. In recent years, with the continuous development of molecular biology technology,intestinal microorganisms have gradually been found to be closely related to human diseases,.which has become a research hotspot. However, the non culturability of some intestinal pathogens limits the research on the pathogenesis of some diseases. Obtaining pure cultures has become the basis and bottleneck to study the interaction, physiological characteristics and pathogenic mechanism of intestinal bacteria. Based on this, this project is intended to study the bacterial culture of human intestinal environment by designing a combination culture method based on MPN pre-culture system and adding the resuscitation-promotion factor (Rpf). .At the same time, the formation and reactivation mechanism of VBNC status of intestinal microorganisms were explored by combining with the macro genomic data of intestinal microorganisms. The aim of this project is to improve the culturability of intestinal microbes,, and to provide theoretical basis and technical support for the culturability of intestinal pathogenic bacteria.
