真核细胞的生命活动依赖细胞器功能的特化和互作,每种细胞器有其特化的功能和特征(如具有不同的pH),通过互作网络来实现物质交换和信息交流,执行不同条件下细胞生命活动的多种生物学过程。细胞内环境和细胞器pH 改变,导致细胞器互作网络的紊乱,与疾病的发生发展密切相关。SNARE蛋白协调细胞器间的识别与融合,是细胞器互作的“分子导航”。目前,活细胞水平还缺乏对多细胞器pH有效监测手段和方法。此外,国内外报道SNARE家族的Syntaxin 12/13可以介导多种细胞器互作,我们预研工作中发现Syntaxin 12/13介导的细胞器互作在多种组织器官中具有重要生理功能,但互作方式、机制和功能目前尚知之甚少。因此本项目拟利用新型高灵敏和宽线性范围pH 探针,用于研究Syntaxin 12/13介导的多细胞器互作,进而理解Syntaxin 12/13介导的多种细胞器互作在细胞、器官、个体水平的生理病理。
英文摘要
Within a eukaryotic cell, there is divided into functional and structural organelles. Each kind of organelle has its special function and characteristic (such as pH), through interaction and mutual coordination to achieve material and information exchange, to form an organelle interaction network, to carry out a variety of biological processes of cell life activities under different conditions. The change of intracellular environment and organelle pH leads to the disorder of organelle interaction network, which is closely related to the occurrence and development of many diseases. At present, there is still lack of an efficient method for monitoring the pH of intracellular organelles; in addition, a variety of organelle interactions mediated by Syntaxin 12/13 have been reported at home and abroad, but the mechanisms and functions of these interactions are still poorly understood. Therefore, this project intends to develop a new pH probe, and used it to study Syntaxin 12/13-mediated multiple organelles interactions, and then understand the functions and roles of the organelles interactions mediated by Syntaxin 12/13 in the modes of cells, organs and animal diseases.
