非编码RNA(non-protein-coding RNA)发挥着重要而广泛的生物学功能,但迄今对其功能的了解还非常有限,对长链非编RNA基因的功能更是知之甚少。水稻基因TL编码一个长非编RNA基因,其反义互补链编码一个MYB转录因子(命名为OsMYB60L1),二者的转录产物可部分互补,形成典型的正义-反义转录本对(Sense-antisense transcript pair)。本项目用已建立的稳定遗传材料(TL-RNAi和OsMYB60L1过量表达转基因植株-两者都使水稻叶片发生螺旋卷曲)开展对水稻长非编码RNA基因功能的深入研究。用链特异转录组测序的方法鉴定差异表达的基因并深度挖掘相关的信息;并用CHIP-seq的方法研究TL对OsMYB60L1的调控机理和分子机制,结合两者构建以非编码RNA基因TL为核心的基因表达调控网络并探讨非编码RNA基因 TL在水稻叶片发育中的调控作用。

Non-protein coding RNAs (npcRNAs) are a class of RNAs that do not encode proteins, but instead their function lies on the RNA molecule, and they are thought to play important and wide biological roles in cells. However, till now much less has been known about the functions of long npcRNAs than about those of small RNAs. TL encodes a putative long non-protein coding RNA, which is antisense to a protein-coding mRNA encoded by OsMYB60L1. Knock-down of TL and over-expression of OsMYB60L1 in the transgenic rice plants both induced twisted leaf blades. Strand-specific RNA sequencing (ssRNA-seq) was used to identify differentially expressed genes. Histone modifications are important epigenetic mechanisms for regulating gene expression. We wanted to use CHIP-seq to explore possible mechanism: TL how to regulate the expression of its antisense gene OsMYB60L1 and explore their regulatory network in leaf development in rice.
