精子发生中阶段特异性的基因表达与调控是最终形成正常精子的分子基础,长非编码RNA(lncRNA)是特异性基因表达的精密调控机器,亦是精子发生的重要调控者。精子发生进程中特别是在精子形成期存在大量特异表达的lncRNA,但对其调控精子发生的具体功能和机制还知之甚少。本研究组前期系统性解析了小鼠精子发生过程中lncRNA的动态表达特征,并通过小鼠体内生精功能筛选和单细胞测序分析等获得了候选lncRNA-1700052I22Rik。该lncRNA特异性表达于圆形精子细胞质。1700052I22Rik基因敲除小鼠精子发生阻滞在圆形精子时期。本项目拟在此基础上,深入研究1700052I22Rik对精子发生进程及雄性小鼠生育的影响,并揭示其通过互作蛋白复合体参与圆形精子细胞变形等精子变形期重要生物学事件的作用机制,有望为阐释精子变形的分子机理以及男性不育相关的的临床实践提供新的科学依据。

Stage-specific gene expression and regulation during spermatogenesis is the molecular basis for the final generation of normal sperm. Long noncoding RNA (lncRNA) is a precise regulator of specific gene expression and an important regulator of spermatogenesis. A large number of lncRNA has been systematically identified in spermatogenesis, specifically during spermiogenesis, but little is known about its specific functions and mechanisms in regulating spermatogenesis. Based on our previous study, we constructed the global lncRNA expression landscape during spermatogenesis and generated candidate lncRNA 1700052I22Rik through co-expression network analysis, in vivo phenotype screening approaches and sing-cell RNA-seq analysis. LncRNA 1700052I22Rik is specifically expressed in round spermatids and is located in the cytoplasm. 1700052I22Rik deletion retarded the development of spermatogenesis, specifically for morphological transformation from round spermatids to elongating spermatids. Based on the existing data, this study will conduct an in-depth study on the effects of 1700052I22Rik on spermatogenesis and the reproduction of male mice, and will reveal the molecular mechanism of its involvement in important biological events during spermiogenesis such as the morphological transformation of round spermatids through the interaction of protein complex. This study will provide scientific basis for elucidating the molecular mechanism of spermiogenesis and the clinical practice of male infertility.
