急性淋巴细胞白血病（ALL）是儿童最常见的恶性肿瘤之一。费城（Ph）染色体阳性是ALL的高危因素。Ph+ ALL患儿易复发和耐药，预后差。使用酪氨酸酶抑制剂（TKI）后大部分可达完全缓解，但仍有30%左右的病人化疗耐药和复发。研究发现Ik6与TKI耐药和预后不良有关。槐耳是一种药用真菌，可促进多种肿瘤细胞凋亡。研究者近期实验发现槐耳可以诱导Ik6和BCR-ABL1共表达的细胞凋亡。因此本研究提出假设：槐耳可通过Ik6、BCR-ABL1双靶点抑制Akt-FoxO1通路增敏Ik6+Ph+ ALL的TKI治疗。课题组和国内外最新研究发现Ik6和BCR-ABL1可通过Akt-FoxO1通路抑制细胞凋亡，本课题将通过体内外及临床实验，进一步研究槐耳对Ik6、BCR-ABL1基因表达水平的影响，来探讨对Ik6+Ph+ALL细胞增殖、凋亡、PTK活性、凋亡通路的作用。

Acute lymphocytic leukemia (ALL) is one of the most common malignant tumors in children.Philadelphia (Ph) chromosome positive is a high risk factor for ALL. Children with Ph + ALL are prone to relapse and drug resistance, and have a poor prognosis. After using tyrosinase inhibitor (TKI), most of them can achieve complete remission, but about 30% of patients still have chemotherapy resistance and relapse. The study found that Ik6 is associated with TKI resistance and poor prognosis. Huaier is a medicinal fungus that can promote apoptosis of various tumor cells. Researchers have recently discovered that Huaier can induce apoptosis of Ik6 and BCR-ABL1 co-expressed cells. Therefore, this study puts forward the hypothesis: Huaier can inhibit the Akt-FoxO1 pathway to sensitize Ik6 + Ph + ALL TKI treatment by dual targets of Ik6 and BCR-ABL1. The research group and the latest research at home and abroad have found that Ik6 and BCR-ABL1 can inhibit apoptosis through the Akt-FoxO1 pathway. This subject will further study the effects of Huaier on the expression levels of Ik6 and BCR-ABL1 genes in vivo and in vitro and clinical experiments. To explore the effects on Ik6 + Ph + ALL cell proliferation, apoptosis, PTK activity, and apoptosis pathway.
