瓦氏雅罗鱼是内蒙古高盐碱湖泊达里湖中仅存的两种经济鱼类之一。近年来,因干旱等自然因素影响使得入湖淡水量锐减,瓦氏雅罗鱼繁殖种群数量显著下降。摸清瓦氏雅罗鱼生殖洄游过程中性腺的发育成熟与盐碱相关水环境因子的关系,解析盐碱环境对达里湖瓦氏雅罗鱼性腺发育的调控机制是突破其人工繁殖技术,保护种群资源亟需解决的首要问题。本项目拟利用形态学、组织学和内分泌学方法研究瓦氏雅罗鱼生殖洄游过程中卵巢发育成熟的调控规律,评估卵巢发育成熟状态与盐碱环境的关系;利用实验生态学方法研究适应不同盐碱环境对其卵巢发育的影响;进一步以HPG为主线,基于Kisspeptin/GPR54系统,运用基因克隆和qPCR技术分析瓦氏雅罗鱼相关激素基因表达对盐碱环境的响应特征,揭示盐碱环境对达里湖瓦氏雅罗鱼生殖洄游过程中卵巢发育的调控机制。该项目的完成将为实现达里湖瓦氏雅罗鱼人工繁殖、保护其种群资源奠定科学的理论基础。

Dali Nor lake is one of the four lakes in Inner Mongolia. It is a typical saline-alkaline lake with high concentrations of carbonate salts (alkalinity up to 60 mmol/L, pH 9.6 and salinity 6 ‰). Amur ide (Leuciscus waleckii) is one of the only two existing economic fish in the Dali Nor Lake. In recently, Drought and other natural causes have caused serious damage to their spawning grounds and led to decline of their naturally breeding population. Break the artificial propagation technology of L. waleckii is an urgent problem to solve for protection of Dali Nor L. waleckii population resource. While resolving the pattern of gonad development and the effect mechanism of saline alkaline environment during spawning migration of L. waleckii is the first step. In this study, we will evaluate the relationships between oocyte development and maturation state of migrating L. waleckii with saline-alkaline environment, and study the effect of L. waleckii adapt to different saline-alkaline environment on ovarian maturation, based on the common fish metrics, histological features of ovary and sex hormone levels. Furthermore, focusing on hypothalamic-pituitary-gonadal axis (HPG) and Kisspeptin/GPR54 system, we use molecular cloning and quantitative real-time PCR techniques to investigate mRNA expression of Kisspeptin/GPR54 system related hormones response to different saline alkaline environment. And clarify the regulation mechanism of saline alkaline environment on ovarian development during spawning migration of the L. waleckii. The achievements will lay on the scientific theory foundation for artificial reproduction and protection of Dali Nor L. waleckii population resource.
