航天员的立位耐力降低和运动能力下降等心血管功能失调症状是失重带来的健康问题之一。文献报道失重条件下血管内皮细胞形态和功能改变是心血管功能失调的重要原因,我们前期发现模拟失重效应促进血管内皮细胞凋亡和迁移等,然而导致这些改变的关键分子仍未阐明。我们通过RNA测序筛选出模拟失重下血管内皮细胞中的差异表达基因RBM3,利用CpG岛预测工具和去甲基化试剂,推测RBM3低表达是由于启动子区高甲基化,而模拟失重下增多的RelB作为转录因子可能促进DNMT3a的表达进而使RBM3启动子区高甲基化,改变细胞功能。本课题拟利用RNAi、染色质免疫共沉淀等技术探讨模拟失重效应下RelB/DNMT3a与RBM3基因启动子区DNA高甲基化的关系,揭示RBM3在失重下血管内皮细胞凋亡、蛋白合成、迁移等功能中的作用。该研究有助于理解失重条件下血管内皮细胞的表观遗传学调控机制,为航天员心血管功能不良的防护提供新思路。

The cardiovascular dysfunction including orthostatic intolerance and disability on physical exercise is one of the health problems for astronauts induced by spaceflight. It is reported that morphological and functional changes play a pivotal role in cardiovascular dysfunction. We have found previously that simulated microgravity induces apoptosis and enhances cell migration. However, the underlying key molecules have not been uncovered. Differentially expressed gene RBM3 was selected through RNA-seq. Prediction of potential CpG island using public online tool and treatment with the DNA demethylating agent indicated that downregulation of RBM3 gene may be due to the hypermethylation of its promoter. The increase of RelB may promote the expression of methyltransferase DNMT3a which may promote the methylation of the RBM3 promoter under simulated microgravity, thus regulating vascular endothelial function. This research aims at detecting the relationship between RelB/DNMT3a and DNA methylation in the RBM3 promoter region under simulated microgravity and clarifying the role of RBM3 expression in apoptosis, protein synthesis and cell migration and so on. This research will deepen the understanding of epigenetic regulations in endothelial functional changes under microgravity and help to provide potential therapeutic actions to protect cardiovascular system in astronauts.
本项目聚焦于失重环境下航天员心血管功能失调的机制研究,特别是血管内皮细胞在模拟失重条件下的功能变化及其分子调控机制。失重环境会导致心血管系统的结构和功能重塑,包括血液转移、血容量减少、心脏功能下降等,进而引发立位耐力不良和运动能力降低等问题。血管内皮细胞在血管功能和结构的调节中起关键作用,而RNA结合基序蛋白3(RBM3)在心血管疾病和血管内皮细胞功能中的作用尚未明确。研究主要围绕两个科学问题展开:①模拟失重是否通过RelB/DNMT3a通路使RBM3启动子区高甲基化从而抑制其表达;②模拟失重下RBM3表达变化如何影响血管功能。通过实验发现,模拟失重条件下,血管内皮细胞中DNMT3a表达增高,导致RBM3启动子区高甲基化,进而抑制RBM3的表达。去甲基化试剂5-氮杂胞苷(5-AZA)可逆转这一效应,提示DNMT3a介导的RBM3启动子区高甲基化是模拟失重导致RBM3表达减少的原因。进一步研究发现,模拟失重下RBM3表达降低导致FN1和WNT5B表达增高,这两种分子与血管内皮细胞功能密切相关。RBM3通过稳定mRNA促进蛋白质合成,其表达降低导致蛋白合成速率下降。过表达RBM3可逆转模拟失重引起的蛋白合成速率下降,而敲低FN1和WNT5B则无此效果,提示RBM3可能通过其他途径影响蛋白合成。研究还发现,模拟失重通过RBM3启动子区高甲基化降低其表达,进而提高FN1的mRNA稳定性,促进内皮细胞迁移。这些结果揭示了RBM3在模拟失重条件下对血管内皮细胞功能的重要调控作用,为失重心血管功能失调的防护提供了新的理论依据和潜在靶点。本项目首次提出RBM3在模拟失重条件下的关键功能,揭示了DNMT3a介导的RBM3启动子区高甲基化及其下游分子FN1和WNT5B在血管内皮细胞功能改变中的作用,深化了对失重致心血管功能失调机制的认识,为航天员健康防护和地面心血管疾病防治提供了新的思路。
