英文摘要
Rett syndrome (RTT) is a severe and progressive neurodevelopmental disorder characterized by the loss of spoken language and motor skills during early childhood. Mutations of the methyl-CpG binding protein 2 (MeCP2) gene are the most prevalent causes of classical RTT cases. However, the precise pathogenic mechanisms of RTT remain uncertain. Previous studies focus on the critical roles of MeCP2 in transcriptional regulation and modulating chromatin structure. However, our current knowledge about its molecular function is not comprehensive. Apart from the role of MeCP2 in transcriptional regulation, its other biological functions have not been fully explored. Recently, we found that the brains from RTT model mice manifest prolonged DNA damage response, indicating it functions in the DNA repair process after DNA damage. Therefore, in the present project, we will explore how MeCP2 is recruited to the sites of DNA damage. We will further investigate the detailed functions and molecular mechanisms of MeCP2 in DNA repair in cells as well as in RTT model mice. These findings will help us for a deeper understanding of the link of DNA repair and the molecular mechanism underlying RTT, and for offering new therapeutic targets for the disease cure.
雷特综合症(Rett syndrome,RTT)是一种严重影响儿童语言、精神和运动能力的神经遗传病。甲基化CpG结合蛋白2(MeCP2)是一种染色质结合蛋白,主要参与转录调控。目前认为该基因突变是导致RTT的主要原因,但RTT病理发生的分子机制尚未明确,并且人们对MeCP2不依赖转录调控功能的了解还不清楚。本项目主要从DNA损伤修复的角度较为深入地研究了MeCP2蛋白的新功能,初步阐明了MeCP2突变导致的DNA修复障碍与RTT发病间的内在联系。本项目基本完成了既定目标。我们发现MeCP2可被迅速招募到DNA双链断裂损伤位点,且该过程依赖于其DNA甲基化结合结构域及由DNMT1介导的DNA甲基化水平;MeCP2缺失会导致DNA损伤修复速率减慢,同源重组和非同源末端连接修复效率降低,基因组不稳定性和细胞对离子辐射的敏感性增加以及修复早期的染色质凝集障碍;分子机制上,我们发现MeCP2可直接结合HDAC1并促进其在DNA双链断裂修复位点的招募,进而影响DNA损伤修复。重要的是,我们还发现MeCP2突变的RTT模型小鼠大脑中DNA损伤水平明显升高,但由于突变的MeCP2不能招募HDAC1,导致DSB修复显著降低。这些结果不仅揭示了MeCP2在维持基因组稳定性方面的新功能,还阐明了MeCP2突变导致的DNA修复受损可能是导致RTT发病的重要原因,从而为了解RTT的发病机制提供了新视角,并为该病治疗手段的研发提供了新思路。
