提高糖、蛋白质等营养物质在植物不同器官的定向运输效率可以促进生长和增产。由胞间连丝介导的共质体运输是植物体内物质转运和信号交流的重要途径。胞间连丝通透性受到动态调节,但其调控机制仍不清楚。申请人前期研究发现PLM基因功能缺失降低鞘脂分子超长链脂肪酸神经酰胺的含量,抑制胞间连丝形态转变,导致胞间连丝通透性和韧皮部同化物卸载增强。然而具体调控机制尚不明确。本项目拟通过细胞生物学、生物化学、分子生物学、组学分析等手段进一步研究 (1)PLM对神经酰胺合成的作用方式 (2)神经酰胺合成对胞间连丝结构组分的影响 (3)PLM-神经酰胺调控胞间连丝通透性与昼夜环境适应。研究结果有望阐明以PLM介导神经酰胺合成为核心的胞间连丝结构与功能调控和环境适应的机制,建立胞间连丝通透性调控研究的基础性工具和前沿性平台,为通过调节共质体运输来促进植物生长和抗逆提供新的思路。
英文摘要
Improving the transport efficiency of sugar, proteins and other nutritions between different parts of plants can promote crop growth and yield. Plasmodesmata-mediated symplastic transport acts as an important way of material and signal exchange in plants. The permeability of plasmodesmata is regulated dynamically, but its mechanism remains unclear. Our previous study proved that the absence of PLM significantly reduced the biosynthesis of ceramide containing very-long-chain fatty acids, a kind of sphingolipids, and inhibited the morphological transformation of plasmodesmata, resulting in enhanced permeability and phloem unloading. However, the regulatory mechanism is still not clear. In this project, we intend to further study (1) the regulation of ceramide synthesis by PLM (2) the alteration of plasmodesmal structural components by PLM-mediatd ceramide synthesis (3) the day-night environment adaptation of PLM-ceramide-mediated plasmodesmal permeability regulation by means of cell biology, biochemistry, molecular biology, proteomics and lipidomics. Our work will further elucidate the mechanism of structural and functional regulation and environmental adaptation of plasmodesmata with PLM-mediated ceramide synthesis as the core, and establish the basic tools and cutting-edge platforms for the study of plasmodesmata permeability regulation. It will also provide a new ideas for promoting plant growth and stress resistance by modulating symplastic transport.
胞间连丝作为植物物质运输和信号交流的关键通道,负责光合同化物、激素、蛋白质、RNA及信号分子等的细胞间转运。提升细胞间连丝介导的源库间糖、蛋白质等营养物质的运输效率,实现其在特定组织和器官的定向高效分配,对促进植物生长和提高作物产量具有重要意义。细胞间连丝的通透性受细胞微环境、发育阶段及环境变化等多种因素的动态调控,然而,其诱导和调控机制尚不明确。细胞间连丝周围沉积的胼胝质影响其通道的开放性,此外我们先前的研究发现PLM基因介导植物鞘脂分子合成及细胞间连丝形态的转变,进而调控特定细胞层共质体的交流,但其细胞和分子调控机制尚需深入探讨。.本研究以韧皮部卸载增强的突变体plm为研究对象,运用分子生物学、蛋白组学、细胞学、生理生化等技术手段,深入探究PLM蛋白的生化功能,分析可能参与细胞间连丝形态调控的蛋白分子,并研究昼夜环境差异对细胞间连丝通透性的影响。研究发现,PLM与同源蛋白SMS和IPCS的生化功能存在差异,参与神经酰胺合成的旁支途径;组学分析揭示了7种细胞间连丝相关蛋白在野生型和突变体中的表达量存在显著差异,荧光融合蛋白转基因植株检测表明AT2G17120、AT3G61560的定位和表达可能受PLM基因调控;PLM基因表现出昼夜表达差异,但不受生物钟调控;叶片细胞间连丝通透性也存在昼夜差异,推测PLM的昼夜表达调控胞间连丝形态差异,进而影响叶片细胞间连丝通透性和物质分子移动。本研究结果进一步拓展了植物鞘脂调控细胞间连丝形态的机制,同时为理解不同环境下植物物质分子的共质体运输机制提供了理论依据和新的研究思路。
