自噬调控与子宫内膜异位症（EMs）密切相关。内质网应激PERK/eIF2α通路在细胞自噬调控起着关键作用，但与EMs的机制尚需进一步研究。前期研究证实蠲痛饮对EMs有显著防治作用，可通过下调TLR4/NF-κB通路，抑制ESC增殖并促进其凋亡。因此我们推测蠲痛饮可能通过内质网应激PERK/eIF2α通路，调节子宫内膜间质细胞(ESC)自噬来阻止EMs发生发展。本项目通过复制EMs大鼠模型和ESC细胞模型，以内质网诱导剂衣霉素和eIF2α磷酸化抑制剂干预ESC细胞，并以蠲痛饮干预，运用病毒转染、RT-PCR、Western Blot、免疫荧光、透射电镜、激光共聚焦显微镜等方法检测其对内质网应激-自噬以及PERK/eIF2α通路相关因子的影响，从PERK/eIF2α信号通路调控内质网应激-自噬深入探讨蠲痛饮对EMs的作用机理，为中医药防治EMs提供依据，也为传承名老中医经验提供新思路。

Autophagy regulation is closely related to endometriosis(EMs).Even though endoplasmic reticulum stress PERK/eIF2α pathway plays a key role in autophagy regulation, it still needs further study on the mechanism with EMs.Previous studies have confirmed that Juantong Decoction has significant preventive and therapeutic effects on EMs. It can inhibit ESC proliferation and promote apoptosis by down-regulating TLR4/NF-κB pathway. Therefore, we infer that Juantong Decoction may prevent the development of EMs by regulating the autophagy of endometrial stromal cells (ESC) through endoplasmic reticulum stress PERK/eIF2α pathway.By replicating EMs rat model and ESC cell model, this project used endoplasmic reticulum inducer chlamycin and eIF2α phosphorylation inhibitor to intervene ESC cells，and then used Juantong Decoction to interpose those cells.We wielded Viral transfection, RT-PCR, Western Blot, immunofluorescence, transmission electron microscopy and laser confocal microscopy methods to detect the effects of endoplasmic reticulum stress-autophagy PERK/eIF2α signaling pathway related factors. We had known that the endoplasmic reticulum stress PERK/eIF2α signaling pathway could regulate ESCs autophagy.Based on this finding，the mechanism of Juantong Decoction on EMs was discussed in depth, which provided a basis for the prevention and treatment of EMs in traditional Chinese medicine, and also provided a new way to inherit the experience of famous and old Chinese medicine.
