动态血液病是指造血系统稳定性丧失导致血细胞数量大幅振荡的一类疾病,该类疾病患者血细胞数量的振荡模式通常十分复杂。本项目通过造血系统动力学模型对四类重要动态血液病的发病机制及治疗方案进行统一研究。为此,本项目首先研究定量刻画复杂振荡特征的方法,建立各类动态血液病振荡模式的识别准则,从而区分振荡的不同来源和产生机制。在此基础上,研究不同血细胞系间的相互作用,将四类动态血液病统一研究,尽可能完整地给出每种参数异常可能导致的动态血液病类型。最后,通过具有多稳态的造血系统动力学模型,研究消除动态血液病血细胞数量振荡的治疗方案。本项目是对四类动态血液病的全面刻画,能加深对造血系统动力学的认识。研究中注重临床数据和动力学分析的结合,能够为进一步的临床研究奠定理论基础。本项目中振荡解特征的刻画和识别需要综合多种数学工具,对数学研究也有推动作用,且其结果可以应用于其他生物振荡系统。
英文摘要
Dynamic hematological diseases are characterized by oscillations in the number of one or all of the circulating blood cells. They are caused by the disorder of hematopoiesis system. The oscillatory patterns are usually complicated in many cases of dynamic hematological diseases. This project systematically studies the dynamical mechanism and therapeutic strategies of four main dynamic hematological diseases. To this end, this project will investigate firstly the methods for precise characteristics of complicated oscillatory solutions and establish the criterion for identification and classification of dynamic hematological diseases. The characterization will also provide a method to identify the origins of dynamic hematological diseases. Based on the characterization of oscillations, this project consolidates the four types of dynamic hematological diseases. This project aims to obtain a list of the possible responses upon variations in each parameter. Finally, this project will also study the therapeutic strategies towards elimination of oscillation given the existence of multistability in the hematological systems. This project aims to a systematic understanding of the four main dynamical hematological diseases with emphasis on the combination of clinical data and mathematical modeling. This project will prompt the understanding of hematopoiesis dynamics as well as provide theoretical evidence for clinical applications. In addition, this project will study the quantitative characterization and identification of complex oscillations with several mathematical methods, which also prompts the progress of mathematics and raises mathematical problems. This method will be applicable in other oscillatory biological systems.
本项目以动态血液病为重点研究造血系统动力学。动态血液病是一类血细胞数量周期性振荡的罕见病。本项目通过改进已有的时滞微分方程模型研究了其中一类动态血液病(周期性血小板减少症)的动力学特征及产生机制。我们得到了血小板有关的动力学参数发生改变时血细胞数量可能出现的各种振荡模式。进一步的结果表明血小板成熟时间和造血干细胞向血小板细胞系的分化率在各种类型的振荡的出现中起着重要作用。分叉分析还揭示这些振荡模式具有不同的动力学起源。这些结果表明血小板细胞系的调控的改变能够引起各种振荡模式,不仅可能导致血小板数量的周期性振荡,还可能引起其他血细胞以及造血干细胞数量的显著振荡。除动态血液病的问题外,本项目还研究了血常规时间序列的分析。给出了一种血常规时间序列数据的特征提取的刻画方法。并在此基础上利用机器学习方法对血常规的时间序列进行分类,得到基于血常规的动态的预警模型。对于疾病的预防、监测等方面具有参考价值。..此外,因为新冠疫情是近两年关注的首要事件,因而本项目也开展了与项目主题有关的新冠疫情的动力学模型方面的研究。我们针对新冠疫情早期的特点,考虑确诊隔离患者和因防疫措施而减少的易感人群两方面因素,对SEIR模型改进得到SEIR-PQ模型。通过对我国主要城市及世界部分国家地区的拟合,验证了模型的合理性。同时,本项目对包括经验函数、统计模型和微分方程模型在内的多种常见传染病模型进行了系统对比和评价。通过池赤信息量、均方根误差和鲁棒性系数,比较分析了各种模型是否能够平衡复杂性和准确性、参数过拟合和欠拟合以及系统鲁棒性和灵敏性这三个冲突准则。我们发现,在训练数据未疫情拐点之后的情况下,涉及的模型均无法得到准确的预测。但是Logsitic函数和Gompertz函数总是分别低估和高估疫情规模;指数增长模型在拐点处会从低估疫情过渡到高估疫情。同时我们也验证了SEIR-PQ模型与我们考虑的其他微分方程模型相比,在上述三方面的具有更好性能表现。这些观察发现可能有助于构建更可靠的预测模型。最后,本项目还从动力学模型的角度对比评估了世界主要国家和地区在疫情早期的政策。
