鹅膏属鞘托鹅膏组真菌能与多种植物形成外生菌根共生关系,具有重要的生态价值。我国该组物种繁丰,全国约73%的物种分布于西南地区。该区丰富的物种多样性可能与青藏高原的几次大规模隆升和横断山脉形成密切相关。在前期研究中,由于所采用的基因片段信息位点有限,且较少关注菌托的显微结构特征,该组下各分支间的系统发育关系不明,我国特别是西南地区的物种多样性认识不足。本项目拟在全国进行广泛采样的基础上,采用浅层基因组测序方法,深度解析鞘托鹅膏组各分支间系统发育关系,利用“形态-分子-生态标准”,强调菌托显微特征,对我国特别是西南地区该组的物种进行精准界定和多样化历史进行研究,探讨其多样化形成与青藏高原隆升和横断山脉形成的关系。本项目对认识我国西南该组的物种起源、演化及扩散过程具有重要科学意义,对该组真菌新资源的开发和利用具有重要现实意义。
英文摘要
Mushrooms of Amanita sect. Vaginatae can form ectomycohrrizal symbioses with various tree species, and are of important ecological value. Species in this section are abundant but difficult to identify, about 73% of the Chinese species are distributed in southwestern China, and the speciation may drive by the uplifts of the Qinghai-Tibet Plateau (QTP) and the formation of Hengduan Mt. Due to the limited gene fragments used and less attention to the structure of volva in many previous studies, the relationships of the species within the section remain unclear, and its species diversity in southwestern China is unsettled. With samples of A. sect. Vaginatae gathered by our team and specimens loaned from domestic and foreign herbaria, this project intends to: 1) construct a deep resolved phylogenetic tree by selecting single-copy orthologous genes through genome skimming; 2) realize accurate species delimitation of China, especially for those in southwestern China, based on "morphological-molecular-ecological criteria" with emphasis on the structure of volva; 3) study the history of species diversification in southwestern China of this section, to elucidate the connection between species diversity and the uplifts of the QTP and the formation of Hengduan Mt. This project is of significant importance in understanding the biodiversity, origin, speciation and diffusion process of this section in southwestern China. Furthermore, this research has important application value in the utilization and protection of mushroom resources.
鞘托鹅膏组真菌能与多种植物形成外生菌根共生关系,具有重要的生态价值。但该组物种繁多,形态可塑性强,且前期研究所纳入分析的基因片段有限,鞘托鹅膏组下包括的主要分支及之间的系统发育亲缘关系尚未明晰,我国该组的物种多样性及其多样化过程仍不清晰。本项目综合系统发育基因组学,多基因系统发育分析,形态学特征,个体发育观察和生态数据,对我国鞘托鹅膏组300余份样品开展了细致研究,发现鞘托鹅膏组位于鹅膏科鹅膏属鹅膏亚属,与橙盖鹅膏组具有较近的亲缘关系,但后者的成员具有菌环和锁状联合。首次利用1016个单拷贝直系同源基因构建了鞘托鹅膏组的系统发育框架,发现其下囊括4个大分支和22个亚分支,并解析了它们之间的系统发育关系。综合多种证据,精准识别了我国该组的46个物种,包括14个新种。同时,发现菌幕残余显微结构为该组物种识别的关键性状,并从个体发育的角度,解析了鹅膏真菌菌幕发育式样及菌幕残余复杂多样性的成因,明确了鞘托鹅膏组菌幕的演化方式。在此基础上,对该组物种的多样化历史进行了分析,发现鞘托鹅膏组与橙盖鹅膏组分化于古新世期间,该组物种在迁移扩散至我国后发生了快速辐射演化。我国西南地区该组物种的快速分化主要发生在始新世晚期至中新世中期,青藏高原形成和横断山脉隆升是导致我国该组物种发生快速辐射演化的关键驱动因子。目前,发表学术论文2篇,在国内外学术交流和会议分享报告2次。在项目执行期间,项目主持人以“优秀人才”引进任副研究员,获得了云南省“兴滇英才支持计划”青年人才头衔,通过硕士生导师遴选,目前正在联合培养指导1名博士生,独立培养指导1名硕士生。本项目对充分认识我国真菌物种多样性,对真菌新资源的利用和保护具有重要的现实意义。
