血管性痴呆(VD)严重威胁人类生命健康,切实改善VD相关性认知障碍是临床亟待解决的难题。既往工作证实海马区神经干细胞(hNSCs)的衰老状态是影响VD患者预后的关键因素,而胚胎干细胞源小细胞外囊泡(ESC-sEVs)可显著改善VD相关性认知障碍,其机制与促进衰老hNSCs神经再生有关。进一步研究发现Gata2及下游Wnt通路可能是调控衰老hNSCs向功能性锥体神经元分化的关键机制,而MEF2可能是ESCs-sEVs发挥调控Gata2的关键因子。据此,本项目拟调控Gata2及Wnt通路以明确其参与调节衰老hNSCs向功能性锥体神经元分化;干预MEF2以证实ESC-sEVs通过转运MEF2上调Gata2而促进VD衰老的hNSCs向功能性锥体神经元分化。最终阐明ESC-sEVs促进衰老的hNSCs分化为功能性锥体神经元治疗VD的作用和具体机制,为VD的治疗研究提供新的理论依据和技术策略。
英文摘要
Vascular dementia (VD) cause a severe threat to public health. However, how to improve the cognitive dysfunction resulted from VD is a critical and urgent question to be solved. Our previous study indicated that hippocampal neural stem cells (hNSCs) senescence played a key role in the prognosis of VD, and treatment with embryonic stem cells-derived small extracellular vesicles (ESC-sEVs) remarkably facilitated cognitive recovery after VD, likely by the promotion of self-renewal and neurogenesis in senescent hNSCs. Furthermore, we also found that Gata2 and the downstream Wnt pathway may be the crucial mechanism to regulate the differentiation of senescent hNSCs into functional pyramidal neurons, and MEF2 may be the key factor for ESC-sEVs to modulate the expression of Gata2. Accordingly, we will use gene reprogramming method to regulate the expression of Gata2 and Wnt signaling pathway to determine that Gata2 modulate the differentiation of senescent hNSCs into functional pyramidal neurons via Wnt signaling pathway, and prove that ESC-sEVs deliver MEF2 to promote the differentiation of senescent hNSCs into functional pyramidal neurons through the up-regulation of Gata2 by means of regulating MEF2 expression in ESC-sEVs. We aim to clarify that the function and specific mechanism of ESC-sEVs on the promotion of senescent hNSCs to differentiate into functional pyramidal neurons for VD treatment, which provide new theoretical foundation and technological strategy to VD therapy.
血管性痴呆(VD)严重威胁人类生命健康,切实改善VD相关性认知障碍是临床亟待解决的难题。既往工作提示海马区神经干细胞(hNSCs)的衰老状态是影响VD患者预后的关键因素,而胚胎干细胞源小细胞外囊泡(ESC-sEVs)可显著改善VD相关性认知障碍,其机制可能与其促进衰老的hNSCs神经再生有关。本项目主要研究了ESC-sEVs促进VD衰老的hNSCs向功能性锥体神经元分化的功能,探讨了Gata2通过调节Wnt通路参与调控衰老的hNSCs向功能性锥体神经元分化,验证了ESC-sEVs通过转运MEF2蛋白,上调Gata2并激活Wnt通路,促进VD后衰老的hNSCs向功能性锥体神经元分化。通过我们的研究,我们在体内和体外实验中均发现经ESC-sEVs干预后的VD小鼠海马区功能性锥体神经元密度明显高于未干预的VD小鼠,锥体神经元的数量、突起长度及相互形成网络连接形态要明显优于对照组,突触传递相关蛋白、有丝分裂后期标记基因和锥体神经元特异性基因的表达水平也显著升高,锥体神经元在高钾离子刺激下具有良好的胞吞和胞吐FM4-64的功能。神经电生理和脑片膜片钳结果均显示ESC-sEV干预可以明显提高VD海马DG区颗粒细胞层的LTP,提高其神经电传导功能。通过 RNA-seq我们发现Gata2在年轻组 hNSCs 和 ESC-sEVs 干预组 hNSCs 中均具最高的表达丰度,Wnt信号通路的差异基因数量最多。通过慢病毒干预hNSCs内Gata2的表达水平,并干预Wnt通路的表达水平,我们发现Gata2敲低后hNSCs分化形成功能性锥体神经元的功能明显下降。使用BML-284能够提高Gata2敲除hNSCs内胞核β-Catenin的表达水平,提高其向锥体神经元分化的能力。而M2782能够抑制Gata2过表达hNSCs内胞核β-Catenin的表达水平,降低其向锥体神经元分化的能力。并且, Gata2的敲低能够抵消ESC-sEVs的促hNSCs向锥体神经元分化的能力,而且拮抗ESC-sEVs内的MEF2能够部分抵消ESC-sEVs的促进VD病程中hNSCs向功能性锥体神经元分化的能力。综上,本项目的研究表明ESC-sEVs可以通过转运MEF2至VD病程中衰老的hNSCs内,从而促进其向功能性锥体神经元分化,促进认知功能的恢复。
