早期康复对卒中后功能恢复有重要意义,但理想的康复策略尚无定论。临床研究发现短时,间歇运动训练可改善卒中后运动功能恢复,与我们的前期研究结果一致。明确短时,间歇运动训练是否是一种理想的卒中早期康复策略,并探索其机制,意义重大。研究证实运动训练可激活mTOR通路,改善运动记忆及学习;PLD2可通过PA激活mTOR通路;PLD2与短暂的脑缺血缺氧密切相关。我们的预实验也发现短时,间歇运动可上调PA、PLD2、mTOR的表达,提升MBP、PSD-95水平,促进功能恢复。故我们提出假说:短时,间歇运动训练是卒中早期理想的康复策略,可以激活PLD2/PA/mTOR通路,改善运动记忆及学习,促进运动功能恢复,改善预后。本项目将使用野生型、PLD2基因敲除小鼠和原代神经元,采用形态学和生物化学方法,明确早期短时,间歇运动训练促进运动功能恢复的作用,并探索其信号传导机制,为脑卒中后精准康复医疗提供理论依据。

Early rehabilitation is of great significance for functional recovery after stroke, but the ideal rehabilitation strategy is inconclusive. Clinical studies have found that short multiple mobilization early after stroke can improve motor function recovery, which is consistent with our previous research results. It is of great significance to determine whether short multiple mobilization is an ideal early rehabilitation strategy for stroke and to explore its mechanism. Studies have demonstrated that motor training can activate mTOR pathway and improve motor memory and learning. PLD2 can activate the mTOR pathway through PA. PLD2 is closely related to transient cerebral ischemia and hypoxia. Our preliminary studies also found that short multiple mobilization could up-regulate the expression of PA, PLD2 and mTOR, improve the level of MBP and PSD-95, and promote functional recovery. Therefore, we hypothesize that short multiple mobilization is an ideal rehabilitation strategy for early stroke, which can improve motor memory and learning, promote motor function recovery and improve prognosis by activating PLD2/PA/mTOR pathway. This project will use wild-type mice, PLD2 gene knockout mice and primary neurons, by using the method of biology and biochemistry studies, molecular and cellular biology, to verify our hypothesis. Our study may provide theoretical basis for the rehabilitation of cerebral apoplexy strategies.
