水稻产量性状的遗传调控机制是水稻功能基因组研究的热点之一。尽管控制产量性状的基因相继被克隆,但是人们仍然对主要产量构成性状的复杂遗传网络不是很清楚。前期对一份单穗粒数显著增加的直立密穗型材料进行QTL检测,本实验室定位到一个新的QTL,并且图位克隆获得了该基因,将其命名为DEP5(Dense and Erect Panicle 5)。本项目以DEP5为诱饵蛋白,通过酵母筛库获得了其互作蛋白SAPK8(osmotic stress/ABA–activated protein kinase 8)。基于已有的研究基础,本项目下一步将利用遗传学方法并结合生物化学和细胞生物学方法,紧密围绕DEP5与SAPK8的遗传互作关系和分子调控机制展开研究。通过本项目的实施,有望加深我们对水稻单穗粒数及相关产量性状调控分子机理的认识,为水稻高产分子设计育种提供理论基础和重要基因资源。
英文摘要
Genetic regulation mechanism of rice yield is one of hot topics in rice functional genomics.Although some grain yield related genes were isolated in recent years , complicated genetic network of grain yield is still poorly understood by us. Previously a new major QTL related to Dense and Erect Panicle was identified. Further this gene was isolated through map-based cloning and we named it DEP5 (Dense and Erect Panicle 5). In this proposal, SAPK8(osmotic stress/ABA–activated protein kinase 8) was picked out through yeast two-hybrid system by using DEP5 as a bait protein.Based on our previous research, our current research work is intended to elucidate the genetic relationship and molecular regulation mechanism between DEP5 and SAPK8 by genetics, biochemistry and cell biology. Taken together, functional analysis of DEP5 and SAPK8 are beneficial to further understanding of regulation mechanism of grain numbers per panicle and grain yield. Meanwhile this work provides a potential tool for improving grain yield in the major crops.
