多数炎症性肠病(IBD)患儿因反复肠道炎症需终生治疗,预后极差。前期研究发现黏膜Th17扩增和转分化是造成肠道炎症的重要诱因(Cell 2019),但在肠道炎症下维持Th17细胞存活的机制不清。文献报道促凋亡蛋白CASP3活化可通过DAP5识别mRNA 5’UTR的IRES元件,启动抗凋亡蛋白翻译促进细胞存活。预实验发现IBD患儿黏膜CD4+T细胞内DAP5、CASP3、抗凋亡蛋白MCL1同时增加;进一步证实Dap5缺失鼠肠炎黏膜Th17含量显著降低, Mcl-1表达降低。据此假设:Th17存活依赖于CASP3启动的DAP5-IRES-MCL1的抗凋亡通路,参与介导IBD发病。将运用Th17细胞条件性DAP5敲除鼠、体外T细胞分化, RIP-RNAseq和移植T细胞肠炎模型等阐述DAP5维持Th17存活机制。该研究将揭示维持Th17细胞存活的新机制,阐述IBD的病理基础并为治疗提供新靶标。

Children with inflammatory bowel diseases (IBD) need lifetime treatment because of recursive gut inflammation. Previous study found massive Th17 expansion and trans-differentiation in the gut mucosa is an important inducement for intestinal inflammation (Cell 2019), while the underlying mechanism for Th17 survival under inflammatory apoptotic environment is unknown. It is reported that activation of pro-apoptotic CASP3 can promote DAP5 to recognize the IRES elements within 5’UTR of mRNAs and selectively initiate anti-apoptotic proteins translation, leading to cell survival. Primary results showed that expressions of CASP3, DAP5 and anti-apoptotic protein MCL1 were co-enhanced in T cells infiltrated in the intestinal mucosa from IBD children. Further experiments showed that Th17 in the inflammatory gut mucosa of Dap5-/- mice had significantly reduced abundance and weaker anti-apoptotic Mcl-1 expression. So we hypothesize that Th17 survival depends on CASP3-initiated DAP5-IRES-MCL1 anti-apoptotic pathway, and plays important roles in IBD pathogenesis. We shall utilize Th17-conditional Dap5 knockout mice, in vitro T cell differentiation, adoptive T cell transfer colitis model, RIP-RNAseq technology to further investigate the molecular mechanism through which DAP5 maintains Th17 survival under apoptotic condition. This study shall strengthen our understanding about Th17 survival mechanisms during IBD progression, providing novel therapeutic target for IBD treatment.
本项目发现非经典翻译起始因子eIF4G2/DAP5通过液-液相分离促进Treg 抗凋亡和增殖相关蛋白的翻译,从而维持Treg 的稳态和功能。条件性敲除Dap5的小鼠外周Treg缺失,并自发发展为致死性的自身免疫反应。结直肠癌队列研究发现Treg内DAP5表达水平与肿瘤内Treg浸润丰度正相关。Dap5单倍体缺失小鼠在稳态和急性炎症中表现正常免疫耐受性,但皮下瘤生长抑制。进一步研究,揭示了Treg生存和适应性依赖于替代性蛋白翻译,表明靶向 Tregs 翻译机制可激发抗肿瘤免疫反应,具有治疗潜力。文章目前在Advanced Science审稿。其他受该经费资助的成果有(1)分离一种新型遗传KDM6A A694T突变,加剧罕见儿童结肠癌的恶性程度并抑制抗肿瘤免疫反应,为肿瘤治疗和预防提供了新思路;(2)发现不成熟的肿瘤相关三级淋巴结构内滤泡外途径的CD11c+T-bet+非典型记忆B细胞(AtM)推动炎癌转变进程,对黏膜上皮肿瘤的早期诊疗具有重要意义;(3)揭示 BACH2在调控二型天然淋巴细胞(ILC2)发育和功能发挥中的重要作用,且BACH2在ILC2 和Th2中呈相反的调控作用,阐明了BACH2在哮喘过敏免疫反应中的多面角色;(4)总结慢性肺病中远端气道损伤的修复缺陷的产生机制及靶向治疗的最新进展,为后续研究指明方向。.共三篇文章分别在《Advanced Science》,《Science Advances》,《Cancer Immunology Research》修稿。已分别在《Oncogene》和《Cell Transplantation》期刊上发表论文各1篇。申请专利1项且已进入实质审查阶段。获批一项国家高层次青年人才项目、联合指导1名博士后出站、指导1名硕士研究生毕业。
