我国香菇产量约占世界总产量的70%,产品以鲜销为主。冷藏是香菇采后贮藏的主要方式,而冷藏过程中的组织软化是导致其商品性下降的主要原因之一。然而,目前香菇采后相关研究多集中于生理水平,对于其采后组织软化的基础研究尚未见系统报道。课题组前期研究发现,多个C2H2型锌指蛋白的基因表达量随贮藏期延长显著升高,且与软化过程呈明显相关性。而C2H2型锌指蛋白是真菌中调控细胞壁β-1,3葡聚糖和几丁质合成的一类重要转录因子,进而会对真菌菌丝的强度与形态结构产生重要影响。为此,本课题拟在筛选出与香菇采后组织软化密切相关的C2H2型锌指蛋白的基础上,进一步对其下游靶基因进行鉴定与互作分析,以期系统解析该类蛋白在香菇采后组织软化中的转录调控网络。研究结果不仅有助于揭示这一与果蔬明显不同的香菇采后组织软化机制,进而为香菇保鲜方法的开发提供理论依据,同时也将为香菇乃至其它食用菌采后品质劣变机制的深入研究提供参考。
英文摘要
The output of Lentinus edodes in China accounts for about 70% of the worldwide, and the fruiting bodies of L. edodes are mainly fresh used. Cold storage is the main way for the postharvest storage of L. edodes, and the softening of the tissue during cold storage is one of the main reasons for the decline of its commodity value. However, the researches on postharvest L. edodes are mainly focused on physiological level, and there is no systematic study on the basic mechanism of postharvest tissue softening. In our previous study, we found that the gene expression of C2H2 zinc finger protein increased significantly with the prolongation of storage period, and had a significant correlation with the postharvest softening of L. edodes fruiting body. Beside this, the regulation of C2H2 type zinc finger proteins in the biosynthesis of cell wall beta -1,3 glucan and chitin in fungi were confirmed in previous study. It implies that the C2H2 zinc finger may regulate the strength and morphological structure of fungal mycelium. In this project, C2H2-type zinc finger proteins involved in the postharvest tissue softening of L. edodes will be selected. The target genes of key C2H2-type zinc finger proteins will be identified, and the regulatory networks of key C2H2-type zinc finger proteins on downstream target genes will be studied systematically. The result of this project has an important guiding significance for the development of postharvest storage methods of L. edodes, and will also provide a reference for revealing the deterioration mechanism of postharvest quality of edible fungi represented by L. edodes.
贮藏过程中的组织软化是导致鲜食香菇商品性状下降的最主要原因,其中涉及的具体机制尚不清楚。本项目明确了香菇采后软化发生特点,建立了香菇采后组织软化模型:采后冷藏期间香菇子实体逐渐软化,表现为组织硬度、咀嚼性和粘弹性等质构指标随贮藏时间延长逐渐下降;细胞壁组分中壳聚糖、几丁质和β-1,3葡聚糖含量,以及几丁质脱乙酰度随贮藏时间延长逐渐下降,同时几丁质酶和β-1,3葡聚糖酶活性在贮藏初始呈现显著上升,但随贮藏时间延长亦呈现下降趋势;透射电镜及扫描电镜结果显示,随着子实体质地软化香菇菌丝组织结构逐渐松散、菌丝间空隙显著增多、细胞结构逐渐被破坏,细胞壁变薄、质膜完整性下降。整体来看香菇采后软化是细胞壁组分降解、机体代谢紊乱和菌丝聚合度下降等因素综合作用的结果。此外,筛选出20个在软化进程中显著差异表达的关键LeC2H2锌指蛋白,其中8个LeC2H2蛋白具有自激活能力;筛选出9个参与细胞壁多糖合成和几丁质修饰相关的基因,以上基因均含有C2H2锌指蛋白顺式作用元件。另外明确了关键LeC2H2锌指蛋白在香菇组织软化进程中的调控机制:LeC2H2-2锌指蛋白能靶向调控LeCHS6基因表达,LeC2H2-27能调控调控LeGNPNAT1基因表达,LeC2H2-29能靶向调控LeHK2和LeGNA2基因表达。本项目建立了香菇采后组织软化模型,鉴定了调控香菇软化进程中细胞壁代谢相关的关键LeC2H2蛋白,明确了关键LeC2H2-细胞壁代谢靶基因互作机制。本项目成果为解析香菇采后组织软化的调控网络奠定了重要基础。
