阻断和干预胃癌前病变（PLGC）对于降低胃癌的发病率和死亡率具有重要意义。课题组在辨证论治基础及精准医疗模式下确立了欣胃颗粒，明显改善PLGC大鼠胃黏膜的病理状态，其作用机制与Wnt/β-catenin激活状态，调控细胞生长、增殖和分化有关。研究表明，以CyclinsCDKs-CKI为核心的G1/S期细胞周期网络调控系统, 是胃癌最常改变的共同通路。申请人研究结果显示，欣胃颗粒能引起PLGC大鼠CyclinD1表达下调，提示其可能通过调控Cyclins-CDKs-CKIs细胞周期分子网络系统，进一步使Wnt/β-catenin激活抑制的生物学事件。因此，本研究拟采用蛋白印迹、RT-PCR、流式细胞及免疫共沉淀等分子生物学手段，研究欣胃颗粒在Cyclins-CDKs为核心的细胞周期分子网络系统中的调控作用及机理，提示欣胃颗粒防治PLGC的生物学机制，为中医药参与PLGC的防治提供实验依据。

Interrupting and intervening precancerous lesions of gastric cancer is important to reduce the morbidity and mortality of gastric cancer. The study group established Xinwei Granules under the basic of differentiation and treatment and the precise medical mode, and improved the pathological state of gastric mucosa in PLGC rats. Its mechanism of action is related to the activation state of Wnt/β-catenin, which regulates cell growth, proliferation and differentiation.The results show that the G1 /S phase cell cycle network, with Cyclins-CDKs-CKI as the core, is the common pathway for gastric cancer. The results showed that Xingwei granules could induce the decrease of Cyclin D1 expression in PLGC rats, suggesting that it could further activate inhibition biological events by regulating Cyclins-CDKs-CKIs cell cycle molecular network system. Therefore, the molecular biology methods of Western blot,Real-time PCR, flow cytometry and immunological Co-precipitation were used to study the regulation and mechanism of Xinwei granules in Cyclins-CDKs cell cycle molecular network. It is suggested that the biological mechanism of Xinwei Granules control PLGC and provides experimental basis for TCM to participate in PLGC control.
