英文摘要
Heart transplantation is the “gold standard” for end-stage heart failure patients. However, donor organ shortage limits the number of heart transplantation. Cold storage is the standard organ preservation method that puts a donor heart into 0 - 4℃ preservation solution until transplantation. It induces a severe decline in organ quality due to anaerobic metabolism and lacks heart functional assessment, resulting in many transplantable donor hearts being discarded. As a novel technology, ex situ heart perfusion maintains the donor heart in a warm beating state and provide an opportunity for heart viability assessment. We have developed a fully automated ex situ heart perfusion instrument and performed over 200 large animals (pig) and three human hearts experiments using our system. In this project, we will develop a non-invasive heart function assessment technique to enable physicians to quantitatively select viable donor hearts. We will study the multi-scale coupling mechanism between physiological characteristics in cell level and cardiac function in large scale level. Through deep learning, a heart transplantation prediction model will be built by extracting heart features from measured parameters. A non-invasive heart function assessment technique will be developed based on multi-sensor information fusion to help physicians to evaluate heart ‘usability’ for transplantation. The research results will provide theoretical and technical support for the high-quality preservation and assessment of donor heart. Ex situ heart perfusion system with non-invasive functional assessment will increase in the number of organs available and reduce the transplantation risk. More qualified donor hearts will help save more lives and families.
