适宜的抽穗期是保证作物高产稳产的重要农艺性状,因此小麦抽穗期新基因的发掘与功能解析具有重要的科学意义和应用价值。在前期工作中,我们从突变体m605中鉴定了一个新的控制小麦抽穗期的基因TaHdm605,并通过图位克隆的方法获得了候选基因。本项目拟对TaHdm605在小麦中调控抽穗期的功能和分子机制进行深入研究,同时筛选优异等位基因。首先,通过对TaHdm605候选基因功能互补、基因编辑及超表达转基因小麦的表型分析,明确TaHdm605对小麦抽穗过程的调控作用。其次,通过TaHdm605互作蛋白筛选、基因表达和RNA-Seq分析,阐明TaHdm605调控小麦抽穗期的分子机制。最后,通过对小麦种质资源分析,发掘TaHdm605优异等位基因。研究结果不仅可以丰富小麦抽穗期调控的理论基础,而且还可以为广适小麦品种的培育提供优异基因资源。
英文摘要
The optimum flowering time is an important agronomic trait for sustainable productivity and high yield potential of crops; therefore, to discover genes regulating heading date and to analyze their functions are of theoretical significance and application value in wheat. In our previous works, a novel gene controlling wheat heading date was identified from the mutant m605 and a candidate gene was isolated by map-based cloning strategy. In this work, we will decipher the effect of TaHdm605 on wheat heading date through series of experiments. Firstly, the role of TaHdm605 on regulating wheat heading date will be determined by analyzing transgenic wheat lines complementary to TaHdm605 and the over-expressing TaHdm605, and knockout lines generated using gene editing technology. Secondly, the molecular mechanism of TaHdm605 regulating wheat heading date will be dissected through interaction protein screening, gene expression analysis and RNA-Seq analysis. Thirdly, the variations of TaHdm605 and their effects on the heading date in wheat will be conducted in wheat germplasm. Through the research above, we hope to provide new information on the genetic and molecular control of heading date in wheat, ultimately to utilize the ideal candidate gene identified for breeding wheat varieties with wide adaptability.
