英文摘要
Androgen sustains prostate cancer development. Disease progression is temporarily postponed by medicine suppressing the steroidogenic pathway, indicating the existence of novel onco-metabolites beyond androgens. Previously, we have found a significant increase of progesterone in patients receiving abiraterone treatment, indicating a switch of cholesterol-steroid metabolism from androgens to progesterone. Here we propose to investigate the clinical significance of progesterone, the related mechanisms of progesterone to stimulate disease progression, and the potential intervention strategy. The potential correlation between plasma concentration of progesterone and treatment response would be evaluated. The transient effect and long term effect of progesterone on prostate cancer cell lines will be investigated and the related mechanisms will be validated in organoids and mouse model. The synthesis pathway of progesterone will be investigated to develop novel strategy for prostate cancer therapy by inhibiting the generation of progesterone. Our results will unveil the significance and novel mechanism of metabolic remodeling in the development of prostate cancer, providing potential biomarker and treatment for disease management.
